摘要
旨在探究成纤维细胞生长因子10(fibroblast growth factors-10, FGF10)基因在贵州黑山羊肌肉及脂肪组织中的表达规律。试验以贵州黑山羊为研究对象,首先克隆了贵州黑山羊FGF10基因CDS区,并对该编码区序列进行了生物信息学分析,其次使用qPCR方法分析1、3、6、9、12、24月龄贵州黑山羊背最长肌、臂三头肌、肱二头肌、皮下脂肪、肾周脂肪和尾脂6个组织中FGF10基因mRNA的表达情况。结果表明,克隆的贵州黑山羊FGF10基因CDS区全长642 bp,共编码213个氨基酸,与山羊和牛同源性较高。FGF10基因在不同月龄贵州黑山羊肌肉和脂肪组织中均有表达,且表达量在幼龄贵州黑山羊肌肉和脂肪组织中较低,随年龄增加FGF10基因表达量呈现逐渐上升的趋势;1月龄时,FGF10基因在皮下脂肪表达量极显著高于其他组织,3月龄和6月龄时在背最长肌和臂三头肌中表达量相对较高,9月龄和12月龄时在皮下脂肪中表达量较高,24月龄时,在肱二头肌中表达量最高,尾脂中最低。
In order to explore the expression pattern of the fibroblast growth factors-10(FCF10)gene in the muscle and adipose tissue of Guizhou Black goats,this experiment took Guizhou Black goats as the experimental subjects.Firstly,the CDS region of the FGF10 gene of Guizhou Black goats was cloned,and the sequence was analyzed bioinformatically.Secondly,qPCR was used to analyze the mRNA expression of the FGF1O gene in the longissimus dorsi,triceps brachii,biceps brachii,subcutaneous fat,perirenal fat and tail fat of Guizhou Black goats aged 1,3,6,9,12 and 24 months.The results showed that the CDS region of the FGF10 gene of Guizhou Black goat was 642 bp in length,encoding 213 amino acids,which had high homology with that of goats and cattle.The results of qPCR showed that the FGF10 gene was expressed in the muscle and adipose tissue of Guizhou Black goats at different months of age,and the expression level was lower in the muscle and adipose tissue of young Guizhou Black goats,and that the expression level of the FCF10 gene showed a gradually increasing trend with the increase of age.The expression level of the FGF10 gene in the subcutaneous fat was significantly higher than that in other tissues of the goats at 1 month of age,relatively higher in the longissimus dorsi and triceps brachi of the goats at 3 and 6 months of age,higher in the subcutaneous fat of those at 9 and 12 months of age,the highest in the biceps brachii and the lowest in the caudal fat of those at 24 months of age.
