基于生物信息学探讨小细胞肺癌的关键基因及潜在治疗药物

摘要 小细胞肺癌(Small Cell Lung Cancer)是一种侵袭性恶性肿瘤,目前尚缺少有效治疗药物。本研究对基因表达综合(Gene Expression Omnibus,GEO)数据库中的GSE149507芯片数据集分析,共获得153个差异基因。通过DAVID进行基因本体论(Gene ontology,GO)发现差异基因主要富集于细胞分裂的生物学过程,KEGG信号通路富集分析提示其主要富集在细胞周期与P53信号通路。利用STRING构建蛋白互作(Protein-protein interaction,PPI)网络,筛选得到一个基因模块,其中最关键的基因为周期蛋白依赖性激酶1(Cyclin-dependent kinases 1,CDK1),提示作用于CDK1的药物可作为治疗小细胞肺癌的潜在药物,最后借助Herb和DrugBank数据库检索作用于CDK1的中药及其单体和化学药物。本研究旨在通过生物信息学挖掘小细胞肺癌发生的分子机制及关键基因,并探讨其潜在治疗药物,为小细胞肺癌的防治提供新思路。 Small cell lung cancer is an aggressive malignancy and there is a lack of effective therapeutic agents.The study analyzed the GSE149507 microarray dataset from the Gene Expression Omnibus(GEO)database and obtained a total of 153 differential genes.Gene ontology(GO)by DAVID revealed that the differential genes were mainly enriched in the biological process of cell division,and KEGG signaling pathway enrichment analysis suggested that they were mainly enriched in the cell cycle and P53 signaling pathways.A protein-protein interaction(PPI)network was constructed using STRING,and a gene module was screened,in which the most critical gene was cyclin-dependent kinases 1(CDK1),suggesting that drugs acting on CDK1 could be potential drugs for the treatment of small cell lung cancer,and finally herbal medicines acting on CDK1 and their monomers and chemical drugs were searched with the help of Herb and DrugBank databases.The study aims to uncover the molecular mechanism and key genes of small cell lung carcinogenesis through bioinformatics,and to explore its potential therapeutic drugs to provide new ideas for the prevention and treatment of small cell lung cancer.
