当归苦参丸止痒功效的初步机制探究

Preliminary mechanism study on the antipruritic effect of Danggui Kushen Pill

目的:探究当归苦参丸改善急性、慢性瘙痒的药效作用,为当归苦参丸的止痒作用及临床用药提供科学依据。方法:采用4-氨基吡啶(4-aminopyridine,4-AP)、右旋糖酐-40(dextran-40)致昆明种小鼠急性瘙痒模型,观察不同剂量当归苦参丸的止痒作用;采用Compound 48/80(C48/80)致小鼠皮肤瘙痒模型,观察不同剂量当归苦参丸对肥大细胞脱颗粒诱发的小鼠瘙痒的干预作用;采用二甲苯致小鼠急性炎症模型,观察不同剂量当归苦参丸的抗炎作用;采用MC903建立特应性皮炎慢性瘙痒模型,观察不同剂量当归苦参丸的抗慢性瘙痒作用。结果:当归苦参丸可改善4-AP和右旋糖酐-40致小鼠急性瘙痒,抑制4-AP小鼠血清中组胺(histamine,HIS)、白三烯B4(leukotriene B4,LTB4)表达;可改善C48/80诱发的瘙痒,抑制肥大细胞脱颗粒和HIS、类胰蛋白酶的表达;可降低二甲苯刺激的小鼠耳肿胀度及提高耳肿胀度抑制率;可改善MC903诱发的特应性皮炎慢性瘙痒,抑制血清中IgE、HIS、TSLP、IL-4、IL-13、IL-33和耳片匀浆中HIS、TSLP、IL-4、IL-13、IL-33的表达水平。结论:当归苦参丸可改善急性、慢性瘙痒,有助于科学性地指导临床用药,提高患者的接受度。

OBJECTIVE To explore the pharmacological effect of Danggui Kushen Pill on improving acute and chronic itching,and to provide scientific basis for the antipruritic effect and clinical medication of Danggui Kushen Pill(DGKSP).METHODS The antipruritic effect of Danggui Kushen Pill at different doses was observed in Kunming mice with acute itching model induced by 4-aminopyridine(4-AP)and dextran-40.To observe the intervention effect of Danggui Kushen Pill with different doses on the pruritus induced by mast cell degranulation in mice,the model of mice skin pruritus induced by Compound 48/80(C48/80)was used.The anti-inflammatory effect of Danggui Kushen Pill with different doses was observed by xylene induced acute inflammation in mice.Using MC903 to establish chronic pruritus model of atopic dermatitis to observe the anti chronic pruritus effect of Danggui Kushen Pill at different doses.RESULTS Danggui Kushen Pill could improve the acute pruritus induced by 4-AP and dextran-40 in mice,and inhibit the expression of histamine(HIS)and leukotriene B4(LTB4)in the serum of 4-AP mice.It can improve the pruritus induced by C48/80,inhibit mast cell degranulation and the expression of HIS and tryptase.It can reduce the ear swelling degree of mice stimulated by xylene and increase the inhibition rate of ear swelling degree.It can improve the chronic pruritus of atopic dermatitis induced by MC903,and inhibit the expression level of IgE,HIS,TSLP,IL-4,IL-13,IL-33 in serum and HIS,TSLP,IL-4,IL-13,IL-33 in ear homogenate.CONCLUSION Danggui Kushen Pill can improve acute and chronic itching,which helps guide clinical medication scientifically and increase patient acceptance.
