生地中活性成分梓醇对秀丽隐杆线虫衰老的影响

摘要 目的探讨梓醇延长秀丽隐杆线虫寿命的作用及其机制。方法以野生型秀丽隐杆线虫(N2)、突变体线虫Daf-2(CF1041)、Daf-16(CF1038)为衰老模型,设置空白组、梓醇低、中、高剂量组(0.025、0.050、0.100 mg/ml梓醇),观察梓醇对线虫寿命的调控作用。通过检测梓醇对N2线虫生殖能力、运动能力、抗急性应激及荧光定量RT-聚合酶链反应(PCR)检测Daf-2、Daf-16、Sod-3及Ctl-1基因表达量,探究梓醇抗衰老作用及机制。结果与空白组比较,梓醇高、中剂量组N2线虫寿命显著延长(P<0.01,P<0.05),突变体Daf-2、Daf-16寿命差异无统计学意义(P>0.05),N2线虫紫外辐射应激、高温热应激、胡桃醌氧化应激及过氧化氢应激条件下生存时长显著延长(P<0.05,P<0.01)。与空白组比较,梓醇高剂量组N2线虫Daf-2 mRNA表达显著下调,Daf-16、Sod-3及Ctl-1 mRNA表达显著上调(P<0.01,P<0.05)。结论梓醇延长了N2线虫正常状态和氧化应激状态下的寿命,没有延长突变体Daf-2和Daf-16线虫寿命,其作用机制可能是通过胰岛素/胰岛素样生长因子(IGF)-1信号通路,促进Daf-16下游相关抗氧化基因表达,提高线虫抗氧化应激能力,从而延长线虫寿命。 Objective To explore the life span extension effect of catalpol and its mechanism using caenorhabditis elegans.Methods Wild-type caenorhabditis elegans(N2),mutant caenorhabditis elegans Daf-2(CF1041),Daf-16(CF1038)were used as aging models.Blank group,low,medium and high dose groups of catalpol(0.025,0.050,0.100 mg/ml catalpol)were set up to observe the regulation effect of catalpol on the lifespan of caenorhabditis elegans.The reproductive ability,locomotion ability,resistance to acute stress of N2 caenorhabditis elegans were detected,and the expressions of Daf-2,Daf-16,Sod-3 and Ctl-1 genes were detected by fluorescence quantitative RT-polymerase chain reaction(PCR)to explore the anti-aging effect of catalpol and its mechanism.Results Compared with the blank group,the lifespan of N2 caenorhabditis elegans in high and medium dose groups of catalpol were significantly extended(P<0.01,P<0.05),and the lifespan of Daf-2 and Daf-16 mutant had no significant difference(P>0.05),the survival time of N2 caenorhabditis elegans was significantly prolonged under ultraviolet radiation stress,high temperature heat stress,juglone oxidative stress and hydrogen peroxide stress(P<0.05,P<0.01).Compare with the blank group,Daf-2 mRNA expression in N2 caenorhabditis elegans was significantly down-regulated,Daf-16,Sod-3 and Ctl-1 mRNA expressions were significantly up-regulated in high dose group of catalpol(P<0.01,P<0.05).Conclusions Catalpol could prolong the lifespan of N2 caenorhabditis elegans under normal and oxidative stress conditions,but not extend the lifespan of mutant Daf-2 and Daf-16 caenorhabditis elegans,the mechanism might be through the insulin/insulin-like growth factor(IGF)-1 signaling pathway,promoting the expression of related antioxidant genes downstream of Daf-16 and improving the antioxidant stress ability of caenorhabditis elegans,thereby prolonging the lifespan of caenorhabditis elegans.
