秋水仙素处理对盐胁迫下青花菜生理生化特性的影响

摘要 【目的】明确秋水仙素处理青花菜种子对提高青花菜耐盐性的影响。【方法】本试验以青花菜种子为材料,使用0.10%、0.20%、0.30%、0.40%秋水仙素处理不同时间(12 h、24 h),以蒸馏水浸泡青花菜种子12 h作为对照,生长7 d后,再以200 mmol/L的NaCl溶液模拟盐胁迫环境,测定青花菜幼苗在盐胁迫下的生理生化特性变化。【结果】200 mmol/L NaCl胁迫可以降低青花菜幼苗过氧化氢酶(CAT)活性、叶片SPAD值和脯氨酸含量,提高超氧化物歧化酶(SOD)、过氧化物酶(POD)活性,丙二醛(MDA)、可溶性糖、可溶性蛋白和硫代葡萄糖苷(硫苷)含量。与CK相比,盐胁迫3 h时,0.30%秋水仙素处理12 h的青花菜幼苗叶片SPAD值、抗氧化酶活性(SOD、POD、CAT)、渗透调节物质(可溶性糖、可溶性蛋白、脯氨酸)和硫苷含量显著提高,MDA含量显著下降。隶属函数综合分析表明,盐胁迫3 h和6 h均使0.30%秋水仙素处理12 h的青花菜幼苗隶属函数值最高,说明适宜浓度秋水仙素处理使青花菜有更好的渗透调节能力、更低的氧化水平和更高的抗氧化能力,从而有助于提高青花菜幼苗的耐盐性。【结论】秋水仙素处理对盐胁迫有一定缓解作用,且以0.30%秋水仙素处理12 h来提高青花菜耐盐性效果最佳。 【Objective】It is to clarify the effect of colchicine treatment on the salt tolerance of broccoli seeds.【Method】In this experiment,broccoli seeds were used as materials,treated with 0.10%,0.20%,0.30%and 0.40%colchicine for different times(12 hours,24 hours)with distilled water soaked broccoli seeds in for 12 hours as a control.After growing for 7 days,the physiological and biochemical characteristics of broccoli under salt stress were measured using 200 mmol/L NaCl solution to simulate the salt stress environment.【Result】The results showed that 200 mmol/L NaCl stress could reduce the CAT activity,photosynthetic pigments,and proline content in broccoli,and increase the activities of SOD,POD,MDA,and soluble sugar,soluble protein and glucosinolates content.Compared with CK,after 3 h of salt stress,the SPAD value,antioxidant enzyme activity(SOD,POD,CAT),osmoregulation substances(soluble sugar,soluble protein,proline),and glucoside content of seedlings treated with 0.30%colchicine for 12 hours significantly increased,while MDA content significantly decreased.The comprehensive analysis of the membership function showed that the membership function values of broccoli seedlings treated with 0.30%colchicine for 12 hours under salt stress for 3 hours and 6 hours are the highest,indicating that appropriate concentration of colchicine treatment makes broccoli have better osmoregulation ability,lower oxidation level,and higher antioxidant capacity,thereby helping to improve the salt tolerance of broccoli seedlings.【Conclusion】Colchicine treatment has a certain mitigation effect on salt stress,and the best effect is to treat broccoli with 0.30%colchicine for 12 hours to improve salt tolerance.
