疏肝健脾益肠方对肝郁脾虚型银屑病小鼠相关炎性细胞因子释放的影响

Effects of Shugan Jianpi Yichang Decoction on the Release of Inflammatory Cytokines in Mice with Psoriasis of Liver Depression and Spleen Deficiency

目的探讨疏肝健脾益肠方对肝郁脾虚型银屑病小鼠相关炎性细胞因子释放的影响。方法40只无特定病原体(SPF)级昆明小鼠(KM小鼠)随机分为正常组、模型组、疏肝健脾益肠方高、中、低剂量组,每组8只。除正常组外,其余各组采用束缚应激、口服醋酸刺激结合咪喹莫特乳膏外涂诱导银屑病小鼠模型;造模结束后,正常组和模型组予以蒸馏水对照,其余各组予以疏肝健脾益肠方进行干预。采用银屑病面积和严重程度指数(PASI)评分评估小鼠背部皮损严重程度;酶联免疫吸附测定法(ELISA法)测定小鼠血清中干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)、白介素-23(IL-23)、白介素-17(IL-17)等相关炎性细胞因子的含量;HE染色检测小鼠皮损组织病理学变化。结果除正常组外,模型组与疏肝健脾益肠方高、中、低剂量组均出现明显的银屑病样皮损,疏肝健脾益肠方组经给药后均有所好转,PASI评分降低(P>0.05);ELISA法结果显示,与正常组比较,模型组血清中IFN-γ、TNF-α、IL-23及IL-17含量均显著升高(P<0.05);与模型组相比,疏肝健脾益肠方高、中、低剂量组小鼠血清中IFN-γ、TNF-α、IL-23及IL-17含量均显著下降(P<0.05)。结论疏肝健脾益肠方可有效治疗银屑病,其机制可能与降低IFN-γ、TNF-α、IL-23及IL-17等相关炎性细胞因子含量有关。

Objective To investigate the effect of Shugan Jianpi Yichang Decoction on the release of inflammatory cytokines in mice with psoriasis of liver depression and spleen deficiency.Methods 40 SPF KM mice were randomly divided into normal group,model group,the high-dose,medium-dose and low-dose group of Shugan Jianpi Yichang decoction,with 8 mice in each group.Except for the normal group,the other groups were treated with binding stress and drink acetic acid stimulation combined with imiquimod cream to induce psoriasis mice model.At the end of modeling,the normal group and model group was given distilled water,and Shugan Jianpi Yichang Decoction was given to other groups for intervention.PASI score was used to evaluate the severity of back lesions in mice.ELISA was used to detect the contents of IFN-γ,TNF-α,IL-23 and IL-17 in serum.HE staining was used to observe the pathological changes of skin tissues.Results Except for the normal group,the other groups had obvious psoriasis-like skin lesions,and therapy groups was improved after treating,and the PASI score decreased(P>0.05).ELISA showed that compared with the normal group,the contents of IFN-γ,TNF-α,IL-23 and IL-17 in serum of psoriasis mice increased significantly(P<0.05);compared with the model group,the contents of IFN-γ,TNF-α,IL-23 and IL-17 in serum decreased significantly(P<0.05).Conclusion Shugan Jianpi Yichang Decoction can effectively treat psoriasis,and its mechanism may be related to reducing the level of inflammatory cytokines such as IFN-γ,TNF-α,IL-23 and IL-17.
