植物病原丝状真菌G蛋白偶联受体的预测方法对比研究

摘要 植物病原丝状真菌G蛋白偶联受体(GPCR)在实现外界信号感知和传递过程中发挥着重要作用。基于GPCR蛋白所具有的典型7个跨膜保守结构域及非典型保守结构域,通过总结前人已报道的候选GPCR的同源比对方法,对其跨膜结构域数量预测方法及跨膜结构蛋白数据库进行对比,明确TOPCONS是目前预测GPCR跨膜结构域的较好方法。然后,选择NCBI数据库中223个不同真菌的GPCR蛋白序列通过TOPCONS和TMHMM进行对比分析,结果显示,2种软件对GPCR跨膜结构域数量的预测结果不尽相同,TOPCONS预测结果的准确度较高;进一步对上述GPCR蛋白序列的氨基酸数目进行分析,明确具有7个跨膜结构域的GPCR蛋白氨基酸数目为200~1406个。该研究可为进一步开展植物病原丝状真菌GPCR蛋白的结构预测及其功能研究提供参考。 G⁃protein⁃coupled receptor(GPCR)of plant pathogenic filamentous fungi plays an important role in external signal per⁃ception and transmission.Based on typical 7 transmembrane conserved domains and other atypical conserved domains of GPCR pro⁃teins,the prediction methods of transmembrane domain number and transmembrane protein databases were compared by summarizing the homologous alignment methods of candidate GPCR reported,and TOPCONS was verified to be a better method to predict the transmembrane domain of GPCR protein at present.Then 223 GPCRs from different fungi were selected from NCBI database for the comparative analysis of TOPCONS and TMHMM.The results showed that the 2 softwares had slightly different prediction results on the number of transmembrane domains,with the higher accuracy of TOPCONS.And further analysis revealed that GPCR proteins with 7 transmembrane domains were composed of 200 to 1406 amino acids.This study can provide references for further structure prediction and functional research on plant pathogenic filamentous fungi GPCR protein.
