基于生物信息学分析LAMB3在口腔鳞状细胞癌中的表达及意义

摘要 目的基于生物信息学方法,通过HPA、TCGA和GEPIA等多个数据库探讨LAMB3在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)中的表达及临床意义。方法HPA数据库分析LAMB3 mRNA和蛋白在不同部位的表达。R软件通过TCGA数据库分析LAMB3 mRNA在OSCC和正常对照样本中的表达差异及与临床病理特征间的关系。GEPIA、TCGA数据库应用Log-rank法行预后分析。BioGRID数据库分析与LAMB3具有交互作用的基因。R软件通过TCGA数据库筛选LAMB3在OSCC中的共表达基因并行富集分析。STRING数据库筛选并构建LAMB3共表达基因的蛋白质互作网络(protein-protein interaction network,PPI network)。结果人正常样本中LAMB3 mRNA在舌部表达最高。LAMB3蛋白在2例正常口腔黏膜样本和2例OSCC样本中均为中度表达。LAMB3 mRNA在OSCC中表达上调,与临床分期、年龄和总生存率相关,与肿瘤分化程度和性别不相关。OSCC中LAMB3的共表达基因中正相关145个、负相关4个。PPI network中具有相互作用关系最多的前10位基因里多为整联蛋白家族成员(ITGA3、ITGA5、ITGA6、ITGB4)。结论LAMB3在OSCC中呈高表达,其高表达与年龄和临床分期相关且预后较差,并且与共表达基因中的整联蛋白在OSCC发生或发展相关的功能与通路的调控机制中关系密切,可为LAMB3为靶点的OSCC临床诊断或治疗提供新的研究方向和依据。 Objective Based on bioinformatics methods,the expression and clinical significance of LAMB3 in oral squamous cell carcinoma(OSCC)were examined.Methods HPA database analyzed the expression of LAMB3 mRNA and protein in different parts.R software analyzed the difference in the expression of LAMB3 mRNA between OSCC and normal control samples and the relationship with clinicopathological characteristics through the TCGA database.Log-rank method was used for prognostic analysis in GEPIA and TCGA database.BioGRID database analyzed genes that interact with LAMB3.R software screened the co-expressed genes of LAMB3 in OSCC through the TCGA database and performed enrichment analysis.STRING database screened and constructed a protein-protein interaction network(PPI network)for LAMB3 co-expressed genes.Results In normal human samples,LAMB3 mRNA was highest in the tongue.LAMB3 protein was moderately expressed in 2 samples of normal oral mucosa and 2 samples of OSCC.LAMB3 mRNA expression was upregulated in OSCC and correlated with clinical stage,age and overall survival,but not with the degree of tumor differentiation and gender.Among the co-expressed genes of LAMB3 in OSCC,145 were positively correlated and 4 were negatively correlated.Most of the top 10 genes with the most interaction relationships in the PPI network were members of the integrin family(ITGA3,ITGA5,ITGA6 and ITGB4).Conclusions LAMB3 was highly expressed in OSCC,and its high expression was related to the age and clinical stage of OSCC and had a poor prognosis.LAMB3 and integrins were closely related to the occurrence or development of OSCC.
