基于指纹图谱及色度分析的不同基原枳实质量差异性研究

Study on the Quality of Aurantii Fructus Immaturus from Different Origins Based on Fingerprinting and Chromaticity Analysis

摘要 目的采用超高效液相色谱(UPLC)指纹图谱、化学模式识别及色度分析,对不同产地的2种基原枳实(酸橙与甜橙)进行质量评价及比较研究。方法以21批酸橙和10批甜橙为研究对象,建立UPLC指纹图谱,并结合聚类分析、主成分分析、偏最小二乘回归分析进行化学模式识别研究;测定31批枳实药材粉末的明度值(L^(*))、黄蓝色值(b^(*))、红绿色值(a^(*)),计算总色度值(E_(ab)^(*)),分析不同基原枳实有效成分含量与其药材粉末色度的相关性。结果酸橙指纹图谱确定24个共有峰,甜橙指纹图谱确定11个共有峰,聚类分析和主成分分析可将31批枳实样品聚为酸橙和甜橙2类,并筛选出11个潜在质量标志物;酸橙E_(ab)^(*)为51.48~68.63,甜橙E_(ab)^(*)为73.33~77.85,药材粉末E_(ab)^(*)与柚皮苷、新橙皮苷、橙皮苷、异橙黄酮和野漆树苷含量均具有显著相关性。结论本研究建立的指纹图谱和含量测定方法简单快速、方便稳定、重复性好,将色度分析与统计学分析相结合可快速鉴别酸橙和甜橙,为不同基原枳实的鉴别和质量评价提供参考。 Objective To perform quality evaluation and comparative study of two original Aurantii Fructus Immaturus of Citrus aurantium L.and Citrus sinensis Osbeck from different habitats through UPLC fingerprint,chemical patterns recognition and colorimetric analysis.Methods The UPLC fingerprints of 21 batches of Citrus aurantium L.and 10 batches of Citrus sinensis Osbeck were established.Combined with clustering analysis,principal component analysis,and partial least squares regression analysis,chemical pattern recognition study was conducted.The lightness value(L^(*)),yellow blue value(b^(*)),red green value(a^(*))of 31 batches of powder were determined,and the total chromaticity value(E_(ab)^(*))was calculated.The correlation between the contents of active ingredients and powder color of different origins of Aurantii Fructus Immaturus was analyzed.Results 24 common peaks were identified in the Citrus aurantium L.fingerprints,and 11 common peaks were identified in the Citrus sinensis Osbeck fingerprints.Clustering analysis and principal component analysis could classify 31 batches of Aurantii Fructus Immaturus into two categories:Citrus aurantium L.and Citrus sinensis Osbeck,and 11 potential quality markers were screened.The E_(ab)^(*) of Citrus aurantium L.was in the range of 51.48‒68.63,and the E_(ab)^(*) of Citrus sinensis Osbeck was in the range of 73.33-77.85.The E_(ab)^(*) of medicinal powder had a significant correlation with the contents of naringin,neohesperidin,hesperidin,isoorange flavone and vernicin.Conclusion The fingerprints and content determination methods established are simple,rapid,convenient,stable,and reproducible,while the chromatographic principle combined with statistical analysis can rapidly identify Citrus aurantium L.and Citrus sinensis Osbeck,which provides a reference for the identification between different origin of Aurantii Fructus Immaturus.
