出入库温度变化对低温贮藏红花种子生活力及耐储性的影响

Effects of Storage Temperature Variation on Viability and Storage Resistance of Carthamus tinctorius L.Seeds Stored at Low Temperature

摘要 如何防止种质生活力意外丧失和延长种子寿命一直是低温种质库种质安全贮藏的研究难点和热点。本研究以红花种子为研究对象,探索出入库温度变化对种子生活力及耐储性的影响。检测不同贮藏温度[中期库(-4±2)℃、长期库(-18±2)℃、超低温冰箱(-75±5)℃]、不同出入库温度变化下,红花种子的生活力指标、耐储性指标及生理生化指标等。结果表明,在不同出入库温度变化下,中期库贮藏种子各指标的差异不显著,长期库贮藏种子的发芽率、可溶性蛋白含量有显著性差异,超低温冰箱贮藏种子的发芽率、发芽指数、可溶性糖、可溶性蛋白、相对电导率、SOD活性、POD活性、CAT活性均有显著性差异。随着贮藏温度的降低,出入库温度变化对红花种子生活力及耐储性的影响越大,红花种子在超低温冰箱贮藏时受出入库温度变化的影响最大,适宜采用快速降温入库和快速升温出库的出入库方法。 How to prevent the accidental loss of germplasm viability and prolong the seed life has been a difficult and hot topic in the research of safe storage of low temperature germplasm bank.In the research,Carthamus tinctorius L.seeds were taken as the research object to explore the effects of temperature changes on seed viability and storage tolerance.The indexes of viability,storage resistance and physiological and biochemical characteristics of Carthamus tinctorius L.seeds were detected under different storage temperatures[(-4±2)℃in mid-term storage,(-18±2)℃in long-term storage,(-75±5)℃in ultra-low temperature refrigerator]and different temperature changes in and out of storage.Results showed that under different storage temperature changes,there were no significant differences in the indexes of safflower seeds stored in mid-term storage,while significant differences in germination rate and soluble protein content of seeds stored in long-term storage,and significant differences in germination rate,germination index,soluble sugar,soluble protein,relative conductivity,SOD activity,POD activity and CAT activity of seeds stored in ultra-low temperature refrigerator.With the decrease of storage temperature,the influence of temperature change on the living ability and storage resistance of Carthamus tinctorius L.seeds was greater.Carthamus tinctorius L.seeds were most affected by temperature change during storage in ultra-low temperature refrigera tor,which was suitable to use the method of rapid cooling into storage and rapid warming out of storage.
