牦牛IGF2基因克隆及生物信息学分析

摘要 为探究牦牛IGF2基因生物学的特征及结构,利用RT-PCR方法克隆了IGF2基因CDS区,并进行了生物信息学分析。结果表明,IGF2基因共有5个开放阅读框,编码区长540 bp,共编码179个氨基酸。该基因编码的蛋白分子式为C864H1377N251O257S9,分子量为19681.51 Da,原子总数2758个,理论等电点9.11,不稳定系数为59.56,总平均亲水性为-0.184,属于亲水性蛋白。IGF2编码蛋白具有73个磷酸化位点且仅存在于细胞质中。通过系统进化树的构建,发现美仁牦牛在进化过程中相对保守。 To explore the biological characteristics and structure of the IGF2 gene in Yak,the CDS region of IGF2 gene was cloned by RT-PCR method,and bioinformatics analysis was performed.The results showed that IGF2 gene had five open reading frames,encoding 540 bp and 179 amino acids.The protein encoded by this gene had molecular formula of C864H1377N251O257S9,molecular weight of 19681.51 Da,total number of atoms of 2758,theoretical isoelectric point of 9.11,instability coefficient of 59.56,and total mean hydrophilicity of-0.184,which belonged to a hydrophilic protein.The IGF2 encoded protein had 73 phosphorylation sites and was present only in the cytoplasm.Through the construction of the phylogenetic tree,it was found that the Meiren yak was relatively conservative during the evolutionary process.
