基于乳酸相关基因肠癌预后模型的建立与分析

摘要 目的寻找新的潜在生物标志物来预测肠癌患者的预后。方法基于TCGA肠癌数据集,分析乳酸相关基因在肠癌中的表达。通过单因素Cox回归分析和LASSO回归分析构建肠癌的乳酸相关基因预后模型。通过CIBERSORT算法评估乳酸相关基因预后评分与肿瘤微环境之间的关系。结果在肠癌中共筛选出78个差异表达的乳酸相关基因用于构建肠癌患者的预后模型。通过Cox回归分析和LASSO回归构建了用于预测肠癌患者预后的乳酸相关基因预后模型。与低风险组比较,高风险组患者总体生存情况较差。低风险组中,T cells CD4 memory resting免疫细胞的浸润情况显著低于高风险组。结论本研究确定了以乳酸相关基因为基础的肠癌预后模型,从而较准确预测肠癌患者的预后,有助于临床医生对肠癌患者进行个体化治疗,进而提高患者生存率。 Objective To find new potential biomarkers to predict the prognosis of colon cancer patients.Methods Based on the TCGA(The Cancer Genome Atlas)colon cancer database,the expression of lactate-related genes in colon cancer was analyzed.A prognostic model of lactate-related genes in colon cancer was constructed by univariate Cox regression analysis and LASSO regression analysis.The relationship between lactate-related gene prognostic score and tumor microenvironment was evaluated by CIBERSORT algorithm.Results A total of 78 differentially expressed lactate-related genes were screened out in colon cancer and used to construct a prognosis model for colon cancer patients.Lactate-related genes were screened out by Cox regression analysis and LASSO regression for the construction of the prognosis risk model of colon cancer.Compared with the low risk group,the overall survival of patients in the high risk group was poorer.In the low risk group,the infiltration of T cells CD4 memory resting immune cells was significantly lower than that in the high risk group.Conclusion The prognostic model for colon cancer based on lactate-related genes can accurately predict the prognosis of colon cancer patients which may be treated individully and improving the survival rate.
