淡水中麦可藻的分离纯化与鉴定

摘要 麦可属(Mychonastes)在淡水中分布广,物种多样性丰富,在生产生物柴油以及净化水质等方面具有潜在的重要价值.但是该类群个体微小,形态简单,需要利用形态以及分子数据对其进行鉴定.从山西省晋中市潇河流域以及运城市新绛县三泉水库采集分离到3株球状绿藻XH-5、XH-13、SQ-9,通过观察其显微结构,分析3个分子标记(18S rRNA、rbc L、tuf A)的系统发育树.结果表明,XH-5、XH-13、SQ-9的18S rRNA序列与麦可属的序列聚为一支,并且3株藻的rbc L和tuf A序列与同球麦可藻(Mychonastes homosphaera)聚为一支,形态上与同球麦可藻的形态相似,基于此,将3株藻鉴定为同球麦可藻.该研究结果可以丰富麦可属在我国的物种多样性和分布状况,促进麦可藻资源的开发与利用. The genus of Mychonastes is widely distributed in fresh water and rich in species diversity,which has potential important value in the production of biodiesel and water purification.However,this group of individuals is small and has a simple morphology,requiring identification using morphological and molecular data.Three spherical green algae XH-5,XH-13,and SQ-9 were collected and isolated from the Xiaohe in Jinzhong City,and the Sanquan Reservoir in Xinjiang County,Yuncheng City,Shanxi Province.By observing their microstructure and analyzing the phylogenetic trees of three molecular markers(18S rRNA,rbc L,tuf A),the results showed that the 18S rRNA sequences of XH-5,XH-13,and SQ-9 clustered into one branch with the sequences of the genus Mychonastes,and the rbc L and tuf A sequences of the three algae strains are clustered into a branch with Myconastes homophaera,which is similar in morphology to Myconastes homophaera.Based on this,the three algae strains are identified as Myconastes homophaera.The results can enrich the species diversity and distribution of Mychonastes in China,and promote the development and utilization of Mychonastes.
