酶解技术对白桦树桑黄有效成分提取工艺研究

摘要 目的以白桦树桑黄为原料,以总多糖和总黄酮含量为指标,在单因素试验基础上,通过正交试验确定白桦桑黄糖化酶的最佳酶解工艺条件。方法使用纤维素酶、糖化酶和淀粉酶对桦树桑黄进行酶解,单因素考察不同酶解方法及酶用量对总多糖、总黄酮含量的影响,并通过L9(34)正交实验优化酶解时间、酶解温度、酶解湿度、酶用量,确定最佳酶解工艺。结果在纤维素酶、糖化酶、淀粉酶发酵作用下,白桦树桑黄的总多糖和总黄酮含量均升高。而在相同的酶活力下,糖化酶使桦树桑黄获得的总多糖及总黄酮综合评价最高。通过正交实验结果可得,白桦树桑黄最佳糖化酶解工艺条件时间为2 d,湿度为55%,加酶量为0.1%(5万μ/g)。结论采用酶解法能极大程度提高白桦树桑黄总多糖、总黄酮的提取率,提供广阔市场需求。 Objective To determine the optimal enzymatic hydrolysis process conditions with orthogonal test by using phellinus igniarius growing on birch tree as raw materials and by using the content of total polysaccharides and flavonoids of betula platyphylla as indexes based on the single-factor experiment.Methods Optimize phellinus linteus were fermented by cellulose,glucoamylase,and amylase,according to the polysaccharide and flavonoids content as index,fermentation time,enzyme solution temperature,humidity,volume of enzyme were elected as influence factors of fermentation and the best optimum fermentation condition was determined with the orthogonal design of L9(34).Results Under the fermentations of cellulase,glycosylase and amylase,the contents of total polysaccharides and flavonoids of birch trees increased.Under the same enzyme activity,the total polysaccharides and total flavonoids obtained from birch trees by glucosidase were the highest.Based on the results of the orthogonal design,the optimum fermentation condition was obtained as enzymolysis time 2 days,enzyme solution temperature at 30℃,55%humidity,and 0.1%(50000μ/g)enzyme dosage.Conclusion The extraction rate of total polysaccharide and total flavone can be greatly improved by enzymatic hydrolysis,which provides a broad market demand.
