摘要
目的:观察雌激素替代治疗对去卵巢糖尿病大鼠心肌组织的影响。方法:构建去卵巢2型糖尿病大鼠模型,在不同条件下继续饲养12周后,对大鼠心肌病理切片行Masson染色,检测左心室质量指数(LVMI)及心肌胶原纤维容积分数(CVF);通过免疫组化法检测大鼠结缔组织生长因子(CTGF)及心肌转化生长因子β1(TGF⁃β1)表达水平;采用电镜观察心肌超微结构。结果:应用雌激素替代治疗后,去卵巢糖尿病大鼠左心室的LVMI、CVF、CTGF、TGF⁃β1均降低(P<0.05),心肌超微结构有所好转。结论:雌激素可以延缓大鼠心肌间质纤维化发生发展,改善心肌超微结构,预防心肌缺血,减少心衰的发生。
Objective:To observe the effect of estrogen replacement therapy on myocardial tissue in ovariectomized rats with diabetes.Methods:The model of ovariectomized rats with type 2 diabetes was established.After 12 weeks of continuous feeding under different conditions,the myocardial pathological sections of the rats were stained with Masson,and the left ventricular mass index(LVMI)and myocardial collagen volume fraction(CVF)were measured.The connective tissue growth factor(CTGF)and myocardial transforming growth factorβ1(TGF⁃β1)of rats were detected by immunohistochemical method.Myocardial ultrastructure was observed using electron microscopy.Results:After estrogen replacement therapy,LVMI,CVF,CTGF,TGF⁃β1 in the left ventricle of ovariectomized rats with diabetes all decreased(P<0.05),and myocardial ultrastructure improved.Conclusion:Estrogen can delay the occurrence and development of myocardial interstitial fibrosis,improve myocardial ultrastructure,prevent myocardial ischemia,and reduce the occurrence of heart failure in rats.
