摘要
通过设计不同的制备载药胶束方法筛选出载药量较高,稳定性较好的载硼替佐米胶束制备工艺。实验采用溶剂挥发法制备载硼替佐米胶束,以反应溶剂、药物质量浓度、溶液pH值等三个因素作为考察因素,确定各因素的最佳参数。结果显示,最适的制备工艺为:反应溶剂为甲醇1 mL+四氢呋喃1 mL的混合溶液,药物质量浓度为5 mg/mL,去离子水溶液pH值为8.5。优化所得的处方工艺可靠、稳定,能够用来制备硼替佐米胶束。
To screen out the preparation process of bortezomib micelle with high drug loading and good stability by designing different preparation methods.In this experiment,bortezomi micelles were prepared by solvent volatilization method.Three factors,such as reaction solvent,drug mass concentration and solution pH value,were used as investigation factors to determine the best parameters of each factor.The optimum preparation process was as follows:the reaction solvent was methanol 1 mL+tetrahydrofuran 1 mL mixed solution,the drug concentration was 5 mg/mL,the pH value of deionized aqueous solution was 8.5.The optimized formulation is reliable and stable,and can be used to prepare bortezomil micelles.
