lncRNA SNHG14介导的miR-223/FBXW7在子宫内膜癌发展中的作用及其临床相关性研究

摘要 目的研究长链非编码RNA(lncRNA)SNHG14在子宫内膜癌发展中的作用及其临床相关性。方法选取2019年1月至2022年1月在海南医学院第二附属医院接受手术治疗的41例子宫内膜癌患者的子宫内膜癌组织和癌旁组织进行研究。采用实时荧光定量聚合酶链反应(qRT-PCR)法检测lncRNA SNHG14、微小RNA(miR)-223和FBXW7表达水平,并探究三者之间关系,分析lncRNA SNHG14与临床病理特征的相关性。将si-lncRNA SNHG14、si-NC、si-miR-223和si-NC转染至HEC-1细胞,采用qRT-PCR法检测lncRNA SNHG14、miR-223和FBXW7表达水平,CCK8法检测细胞增殖水平,Transwell法检测细胞迁移水平;通过生物信息学预测和荧光素酶报告基因实验检测lncRNA SNHG14和miR-223、miR-223和FBXW7的靶向性。结果与癌旁组织相比,子宫内膜癌组织中lncRNA SNHG14表达水平降低,lncRNA SNHG14和miR-223表达、miR-223和FBXW7表达呈负相关,lncRNA SNHG14和FBXW7表达呈正相关。lncRNA SNHG14低表达与肿瘤分期和淋巴结转移相关。与si-NC组相比,si-lncRNA SNHG14组lncRNA SNHG14表达降低,miR-223表达升高,细胞增殖水平和迁移水平增加。lncRNA SNHG14靶向miR-223。与si-NC组相比,si-miR-223组miR-223表达降低,FBXW7表达升高,细胞增殖水平和迁移水平降低。miR-223靶向FBXW7。结论lncRNA SNHG14表达水平与子宫内膜癌临床恶化指标呈负相关,其通过miR-223/FBXW7轴抑制子宫内膜癌细胞的增殖和迁移。 Objective To investigate the role of long noncoding RNA(lncRNA)SNHG14 in the development of endometrial cancer and its clinical relevance.Methods Cancer tissues and adjacent tissues of 41 patients with endometrial cancer undergoing surgical treatment were collected from the Second Affiliated Hospital of Hainan Medical College from January 2019 to January 2022.Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression levels of lncRNA SNHG14,microRNA(miR)-223 and FBXW7.The relationships among these three were investigated and the correlation between lncRNA SNHG14 and clinical pathological features were analyzed.Si-lncRNA SNHG14,si-NC,si-miR-223 and si-NC were transfected into HEC-1 cells.The expression levels of lncRNA SNHG14,miR-223 and FBXW7 were detected by qRT-PCR,cell proliferation was detected by CCK8,and cell migration was detected by Transwell.Bioinformatics prediction and luciferase reporter assay were used to detect the targeting of lncRNA SNHG14 and miR-223,miR-223 and FBXW7.Results Compared with adjacent tissues,the expression level of lncRNA SNHG14 was decreased in endometrial cancer tissues,lncRNA SNHG14 and the expression of miR-223,miR-223 and FBXW7 were negatively correlated,while lncRNA SNHG14 and FBXW7 were positively correlated,and the low expression of lncRNA SNHG14 was associated with tumor stage and lymph node metastasis.Compared with si-NC group,the expression of lncRNA SNHG14 was decreased,the expression of miR-223 was increased,and the cell proliferation and migration levels were increased in si-lncRNA SNHG14 group,and lncRNA SNHG14 targeted miR-223.Compared with si-NC group,the expression of miR-223 was decreased,the expression of FBXW7 was increased,and the level of cell proliferation and migration were decreased in si-miR-223 group,and miR-223 targeted FBXW7.Conclusions The expression level of lncRNA SNHG14 is negatively correlated with the clinical deterioration indicators of endometrial cancer,and it inhibits the proliferation and migration of endometrial cancer cells through miR-223/FBXW7 axis.
