秦皮乙素对大鼠心肌组织抗氧化能力的影响

摘要 目的:探讨秦皮乙素(esculetin,ESC)对多柔比星(doxorubicin,DOX)诱导心肌细胞氧化应激的影响。方法:将SD大鼠分为对照组、DOX组、ESC低、高剂量+DOX组,检测各组大鼠心肌组织中SOD、MDA及血清心肌酶变化;通过Masson染色,比较各组心肌组织中胶原纤维的量的变化;Western blot法检测抗氧化相关蛋白的表达。结果:ESC可升高DOX诱导后心肌组织中SOD水平,同时降低由DOX所致心肌酶及MDA升高(P<0.05);DOX干预后心肌组织中胶原纤维明显增多,而ESC预处理后的心肌组织胶原纤维明显减少;ESC可降低由DOX引起P53含量的增多(P<0.05),并上调心肌组织中的抗氧化酶HO-1(P<0.05)、MDM2(P<0.05)和Nrf2(P<0.05)的表达。结论:秦皮乙素可改善多柔比星诱导的心肌组织氧化应激损伤,并对心肌组织起保护作用。 Objective:To investigate the effect of esculetin(ESC)on the oxidative stress induced by doxorubicin(DOX)in cardiomyocytes.Methods:SD rats were divided into control group,DOX group,ESC low dose and high dose+DOX group.The serum myocardial enzymes and SOD,MDA levels in myocardial tissue were detected in each group.The changes of collagen fibers in myocardial tissue were compared by Masson staining.The expression of related proteins in each group was detected by Western blot.Results:ESC can increase the SOD level and decrease the increase of myocardial enzyme and MDA caused by DOX(P<0.05);after DOX intervention,the collagen fiber in myocardial tissue increased significantly,while the collagen fiber in myocardial tissue pre-treated by ESC decreased significantly;ESC can reduce the increase of p53 content caused by DOX(P<0.05),and up-regulate the antioxidant enzyme HO-1(P<0.05),MDM2(P<0.05)and Nrf2(P<0.05).Conclusion:Ganoderma polysaccharide can improve the oxidative stress injury of myocardial tissue induced by doxorubicin and play a protective role in myocardial tissue.
