摘要
目的探讨黄芩素(BAI)对卵巢癌Skov3细胞增殖与凋亡的影响及可能机制。方法使用不同浓度的BAI处理卵巢癌Skov3细胞,以MTT法检测Skov3细胞增殖,以克隆形成实验检测Skov3细胞克隆能力,以流式细胞术检测Skov3细胞凋亡,以Western blot法检测细胞凋亡相关蛋白以及核因子κB/B淋巴细胞瘤2(NF-κB/Bcl-2)信号通路相关蛋白,以PCR法检测细胞NF-κB/Bcl-2通路相关m RNA,以免疫荧光实验观察细胞中NF-κB p65表达。结果对比对照组,随着BAI药物浓度的增加,卵巢癌Skov3细胞增殖受到抑制,细胞凋亡率显著上升,NF-κB/Bcl-2通路相关蛋白p NF-κB p65、p IκB-α、Bcl-2及相关m RNA NF-κb、Bcl-2表达明显下降,ⅠκB-α表达明显升高。结论BAI可抑制卵巢癌SKOV3细胞的增殖,促进其凋亡,可能与抑制NF-κB/Bcl-2通路有关。
Objective To investigate the effect and possible mechanism of baicalein(BAI)on the proliferation and apoptosis of ovarian cancer Skov3 cells.Methods Different concentrations of BAI were used to treat ovarian cancer Skov3 cells.The effect of BAI on the proliferation of ovarian cancer Skov3 cells was detected by the MTT method.The effect of BAI on the cloning ability and apoptosis were detected by the clone formation experiment and flow cytometry respectively.The expression of apoptosis-related proteins and NF-κB/Bcl-2 pathway related proteins and related mRNA were detected by western blot and PCR respectively.The changes of NF-κB p65 expression was detected by immunofluorescence experiment.Results Compared with the control group,as the concentration of BAI increases,the proliferation of ovarian cancer Skov3 cells was inhibited,the apoptosis rate of ovarian cancer Skov3 cells increased significantly,the expression of NF-κB/Bcl-2 pathway related proteins pNF-κB p65,pIκB-α,Bcl-2 and mRNAs NF-κB Bcl-2 was significantly lower,and the expression ofⅠκB-αwas significantly higher.Conclusion BAI can inhibit the proliferation of ovarian cancer Skov3 cells and promote apoptosis,which may be related to inhibit the NF-κB/Bcl-2 pathway.
