等位菜豆凝集素含量与SSR标记的关联分析 被引量:1

摘要 菜豆豆荚中的凝集素(Phytohaem agglutinin,PHA)是其食用过程中致使人中毒的主要因素。为探究菜豆种质资源中PHA含量差异,挖掘与PHA含量显著相关的功能标记,测定了64份菜豆品种两年中PHA含量变化,设计并筛选出60对SSR(Simple sequence repeats)标记,对菜豆种质进行群体遗传结构分析,并利用TASSEL 2.1软件中的一般线性模型(Generalized linear model,GLM)对菜豆凝集素的含量性状进行关联分析。结果表明,64份菜豆品种中,2018年凝集素含量为0.015~1.564 mg·g^(-1),平均值为0.721 mg·g^(-1),2019年凝集素含量为0.041~1.850 mg·g^(-1),平均值为0.737 mg·g^(-1);60对SSR标记共检测到219个等位位点;遗传相似系数(Genetic similar cofficient,GS)为0.694时,64份菜豆种质可聚为2个类群。关联分析显示,两个等位位点与菜豆凝集素含量性状显著相关(P<0.05),对表型的变异贡献率分别为15.23%和15.05%。 Phytohaem agglutinin(PHA)in the pods of common bean is the main factor of poisoning.In order to explore the difference of PHA content in common bean germplasm resources and identify the markers regulating PHA content,the changes of PHA content were measured in 64 common bean cultivars in two years,60 pairs of SSR(Simple sequence repeats)markers were designed and screened to analyze the genetic structure of common bean population.The association of PHA content in common bean and SSR markers was analyzed using the generalized linear model(GLM)of software TASSEL 2.1.The results showed that the PHA content of 64 common bean varieties was 0.015~1.564 mg·g^(-1)with an average of 0.721 mg·g^(-1) in 2018.The PHA content was 0.041-1.850 mg·g^(-1)with an average of 0.737 mg·g^(-1)in 2019.219 alleles were detected by 60 pairs of SSR markers.When the genetic similar coefficient(GS)was set as 0.694,64 accessions could be clustered into two groups.The association analysis showed that the two alleles were significantly correlated with the PHA content of common bean(P<0.05),and their contribution rates to phenotypic variation were 15.23%and 15.05%,respectively.
