P-PARV4全基因组克隆及序列分析

Sequence analysis of complete genome of porcine parvovirus 4

摘要 为研究广西地区猪hokovirus(PHoV)又称猪PARV4(P-PARV4)毒株流行情况和遗传进化特征,对采自广西地区猪组织样品进行检测,扩增出其中1株全基因序列,命名为YL172(GenBank登录号:KX827773)。感染率调查结果显示,广西地区成年猪P-PARV4感染率为95.0%。同源性分析结果显示,P-PARV4 YL172株全基因组与喀麦隆、欧洲、美国和中国参考株同源性为96.1%~99.3%;YL172株NS1基因与参考株同源性为97.0%~99.5%;YL172株VP2基因与参考株同源性为93.4%~99.1%。VP2遗传进化分析结果显示,P-PARV4分离株存在2个亚群,YL172株与中国参考株GX1、SH1、JSNJ处在同一进化分支,而美国参考株US-HK133与HK1则处在另一进化分支。 To evaluate theprevalence and genetic evolution of porcine parvovirus 4(P-PARV4)strain in Guangxi Province,pig tissue samples from Guangxi region was detected,the whole genome sequence of one strain was amplified,the strain was named P-PARV4 YL172(GenBank accession number:KX827773).The results of infection rate survey showed that the infection rate of P-PARV4 in adult pigs in Guangxi area was 95.0%.Homology analysis showed that the homology of P-PARV4 YL172 strain with reference strains from Cameroon,Europe,USA and China was 96.1%-99.3%.The homology of NS1 was 97.0%-99.5%,the homology of VP2 was 93.4%-99.1%between reference strain and YL172 strain.The phylogenetic analysis of VP2 showed there were two subgroups of P-PARV4 isolates,YL172 strain and China isolates GX1,SH1,JSNJ were in the same evolutionary branch,while US isolates US-HK133 and HK1 were in another evolutionary branch.This study provided a scientific basis for the prevention and control of P-PARV4,and has significant public health importance.
