自然感染绵羊肺腺瘤病毒病肺组织中凋亡相关因子的表达分析 被引量:1

Expression analysis of apoptosis-related factors in lung tissues of sheep naturally infected with Jaagsiekte sheep retrovirus

摘要 绵羊肺腺瘤(ovine pulmonary adenocarcinoma,OPA)是一种由绵羊肺腺瘤病毒(Jaagsiekte sheep retrovirus,JSRV)引起的慢性、接触传染性肺脏肿瘤疾病。为了探究细胞凋亡在OPA发展过程中的作用,本研究利用实时荧光定量PCR、蛋白免疫印迹等方法检测了凋亡相关基因Bcl-2、Bax、Caspase-3及其对应的凋亡相关蛋白Bcl-2、Bax、Caspase-3在健康和自然感染JSRV绵羊肺组织中的表达情况。结果显示:与对照组相比,OPA肺组织中Bcl-2基因和蛋白表达量均显著上调,而Bax和Caspase-3基因和蛋白表达量均显著下调。此外,免疫组织化学显示,Bcl-2、Bax、Caspase-3主要表达于腺瘤灶内增生的Ⅱ型肺泡上皮细胞中。结果表明,细胞凋亡在OPA肺组织中的活动受到抑制,提示绵羊肺腺瘤病毒可能通过调节细胞凋亡活动影响OPA的发生发展。 Ovine pulmonary adenocarcinoma(OPA)is a chronic and contagious pulmonary tumor disease caused by Jaagsiekte sheep retrovirus(JSRV).In order to explore the role of apoptosis in the development of OPA,the mRNA and protein expression of apoptosis-related factors Bcl-2,Bax and Caspase-3 were analyzed by qPCR and Western blot in lung tissues of healthy sheep and sheep lung tissues naturally infected with JSRV.The results showed that,comparing to the control group,the mRNA and protein expression of Bcl-2 were significantly increased in OPA group,while the expression of Bax and Caspase-3 were significantly decreased.In addition,immunohistochemistry results showed that Bcl-2,Bax and Caspase-3 were mainly located in proliferated typeⅡalveolar epithelial cells in adenomas.In conclusion,these results revealed that apoptosis activity was inhibited in OPA lung tissues,suggesting that JSRV infection may affect the occurrence and development of OPA by regulating apoptosis.
