不同浓度臭氧化生理盐水对家兔膝关节软骨的影响

Effects of different ozonated saline concentrations on rabbit knee cartilage

摘要 目的 通过研究不同浓度臭氧化生理盐水对家兔膝关节软骨的影响,探索臭氧水运用的安全性。方法 将20只新西兰家兔按不同的臭氧化生理盐水浓度分为4组,即Ⅰ组(30μg/ml),Ⅱ组(35μg/ml),Ⅲ组(40μg/ml),Ⅳ组(45μg/ml)。分2次分别对家兔膝关节腔内注射臭氧化生理盐水,注射间隔时间为7天。第二次注射7天后进行病理活检及病理检查。结果 Ⅰ组中3例正常,2例出现轻度病理损伤。Ⅱ组、Ⅲ组、Ⅳ组均导致关节软骨不同类型的病理改变。参照骨关节炎研究国际学会(Osteoarthritis Research Society International,OARSI)骨关节炎病理评价系统中的软骨组织病理分级标准,病理分级结果如下:Ⅰ组0级3例,1级2例;Ⅱ组1级3例,2级1例,3级1例;Ⅲ组3级4例;Ⅳ组2级1例,3级3例。运用Spearman等级资料相关关系分析病理分级,结果示r=0.515,P=0.002,差异有统计学意义(P <0.05)。结论 臭氧化生理盐水浓度> 30μg/ml可能对正常家兔膝关节软骨造成损伤,且随着臭氧化生理盐水浓度的递增,引起膝关节损伤的程度也会增加。 Objective To explore safety of ozonated saline by studying the effects of different concentrations of ozonated saline on rabbit knee cartilage.Methods Twenty New Zealand rabbits were divided into four groups according to the concentration of ozonated saline,group Ⅰ( 30 μg/ml),group Ⅱ( 35 μg/ml),group Ⅲ( 40 μg/ml)and group Ⅳ( 45 μg/ml).The rabbits were injected with ozonated saline into the knee joint cavity twice in 7-day interval.Pathological biopsy and examination were performed the 7th day after the second injection.Results In group Ⅰ,3 cases were normal,while mild pathological injury occurred in 2 cases.Articular cartilage changes of different pathological types in group Ⅱ,Ⅲ and Ⅳ were observed.Based on the pathological grading standard of cartilage tissue in the pathological evaluation system of OARSI osteoarthritis:3 cases of grade 0 and 2 cases of grade 1 in group Ⅰ;3 cases of grade 1,1 case of grade 2 and 1 case of grade 3 in group Ⅱ;4 cases of grade 3 in group Ⅲ;1 case of grade 2 and 3 cases of grade 3 in group Ⅳ.Using Spearman grade data to analyze the pathological grade,differences were statistically significant( r = 0.515,P = 0.002)( P<0.05).Conclusions The concentration of ozonated normal saline ≥30 μg/ml may cause damage to the knee cartilage of normal rabbits,and the knee injury will be aggravated with the increase of the concentration of ozonated normal saline.
