micro-CT观察晚期氧化蛋白产物对兔骨关节炎软骨下骨的影响

Quantification of subchondral bone changes following advanced oxidation protein product treatment in a rabbit osteoarthritis model using micro-CT

摘要 目的 利用micro-CT观察外源性晚期氧化蛋白产物(advanced oxidation protein products,AOPPs)对兔骨关节炎软骨下骨骨重塑以及三维结构的影响。方法 将24只雄性新西兰大白兔随机平均分为3组;晚期氧化蛋白产物组(AOPPs组)造模后于双侧膝关节内注射200μg/ml AOPPs 1 ml;磷酸盐缓冲液(phosphate buffered saline,PBS)组在造模后注射等剂量磷酸盐缓冲液;假手术组(Sham组)显露前交叉韧带后缝合切口。干预8周后处死动物并取胫骨标本,对胫骨平台下软骨下骨部分行组织学切片染色与micro-CT扫描,获得相关参数并进行统计学分析。结果 Sham组与PBS组的骨体积分数均较AOPPs组高(P <0.01)。Sham组骨小梁数目高于PBS组(P <0.01)。Sham组与PBS组的骨小梁分离度均低于AOPPs组(P <0.01,P <0.05)。Sham组的骨小梁厚度与连接密度均较AOPPs组高(P <0.05)。骨小梁数目、骨小梁厚度、骨小梁分离度以及连接密度均与骨体积分数存在显著相关关系(P <0.05),且其中骨小梁数目、骨小梁分离度与连接密度为骨体积分数改变的主要因素。结论 外源性AOPPs对兔骨关节炎的软骨下骨产生有明显的影响,加速了软骨下松质骨的骨吸收过程。 Objective To observe the effect of exogenous advanced oxidation protein product on subchondral bone remodeling and three-dimensional structure in a rabbit model of osteoarthritis by Micro-CT.Methods The osteoarthritis model was created by anterior cruciate ligament transection and medial meniscus resection.All 24 male New Zealand white rabbits were randomly divided into 3 groups.The advanced oxidation protein product group( AOPPs group) was injected with 200 μg/ml AOPPs 1ml inside the knee joint after molding surgery;the phosphate buffer group( PBS group) was injected with 1ml phosphate buffer after molding surgery;In sham-operated group( Sham group),the anterior cruciate ligament was just exposed without transection,and then the incision was sutured.Eight weeks after intervention,animals were sacrificed and tibial samples were taken.Histological sections of the subchondral bone of the tibial plateau were stained.Micro-CT was applied to obtain relevant parameters and statistical analysis.Results The bone volume fraction of the Sham and PBS group was higher than the AOPPs group( P<0.01).The number of trabecular bone was higher in the Sham group than that in the AOPPs group( P<0.01).The separation of trabecular bone between the Sham and PBS group was lower than that in the AOPPs group( P<0.01,P<0.05).The trabecular bone thickness and connection density in the Sham group were higher than those in the AOPPs group( P<0.05).The number of trabecular bone,trabecular thickness,trabecular separation and connection density were significantly correlated with bone volume fraction( P<0.05).The number of trabecular bone,trabecular separation and connection density were the main factors for the change of bone volume fraction.Conclusions Exogenous AOPPs accelerates the bone resorption process in subchondral cancellous bone in a rabbit model of osteoarthritis.
