灯盏花素对铅中毒大鼠心肌细胞氧化应激损伤的影响 被引量:1

The influences of breviscapine on oxidative stress in cardiomyocytes of lead-poisoning rats

摘要 目的研究灯盏花素(Bre)对铅中毒大鼠心肌细胞氧化应激损伤的影响。方法选择60只健康成年SD大鼠随机分为对照组、模型组,及Bre干预组12.5 mg/(kg·d)组、25 mg/(kg·d)组和50mg/(kg·d)组,每组12只。后4组用2%醋酸铅灌胃染毒,构建铅中毒大鼠模型,造模成功后干预组灌胃Bre,对照组及模型组用同剂量蒸馏水灌胃,共计处理时间14周。HE染色观察大鼠心肌细胞病理变化,ELISA法测定大鼠氧化应激指标。结果对照组心肌细胞排列正常,模型组心肌细胞排列紊乱,心肌间质可见纤维素样物沉积及淋巴细胞浸润;Bre干预组心肌间质纤维素样物沉积及淋巴细胞浸润较模型组减少。模型组大鼠氧化指标MDA、ROS分别为(3.96±0.69)nmol/mL和(324.45±57.45)U/mL,较对照组MDA(2.92±0.79)nmol/mL、ROS(193.71±76.97)U/mL增高(F=4.72、6.59,P<0.01);抗氧化指标SOD、CAT、GSH分别为(18.19±4.84)U/mL、(9.3±2.07)U/mL、(113.63±32,80)mmol/L,较对照组SOD(36.99±12.66)U/mL、CAT(20.02±4.87)U/mL、GSH(212.66±99.36)mmol/L减低,(F=7.77、12.67、3.94,P<0.01)。Bre干预组氧化指标MDA、ROS含量较模型组减低,抗氧化指标SOD和CAT活力、GSH含量及GSH相关酶GPx、GSR、GST活力较模型组升高,其中以Bre25 mg/(kg·d)组和50 mg/(kg·d)组作用较为明显(F=4.72、6.59、7.77、12.67、3.94、3.18、4.86、4.48,P<0.01)。结论灯盏花素可通过抑制氧化应激反应减轻铅中毒大鼠心肌细胞的损伤。 Objective To study the effect of breviscapine(Bre)on oxidative stress injury of cardiomyocytes in rats with lead poisoning.Methods A total of 60 healthy adult SD rats were selected and randomly divided into five goups(12 per group):the control group,the model group and three Bre intervention groups at 2.5 mg/(kg·d),25 mg/(kg·d),and 50 mg/(kg·d).The last four groups were poisoned with 2%lead acetate though intragastric administration,establishing the lead poisoning rat model.After the model was successfully established,the intervention group was given Bre by gavage,and the control and model groups were given the same dose of distilled water by gavage.The total processing duration was 14 weeks.HE staining was used to observe the pathological changes of rat cardiomyocytes,and ELISA was used to determine the oxidative stress index in rats.Results The arrangement of myocardial cells in the control group was normal,while the arrangement of myocardial cells in the model group was disordered,and there were cellulosic deposits and lymphocyte infiltration in the myocardial interstitium;Compared with the model group,the deposition of myocardial interstitial cellulose and lymphocyte infiltration in the Bre intervention group decreased.MDA and ROS levels in the model group were(3.96±0.69)nmol/mL and(324.45±57.45)U/mL,respectively,which were significantly higher than those in the control group MDA(2.92±0.79)nmol/mL and ROS(193.71±76.97)U/mL(F=4.72,6.59,P<0.01);The antioxidant indexes of SOD,CAT,and GSH were(18.19±4.84)U/mL,(9.3±2.07)U/mL,(113.63±32,80)mmol/L,respectively,which were significantly lower than those of the control group(36.99±12.66)U/mL,CAT(20.02±4.87)U/mL,and GSH(212.66±99.36)mmol/L(F=7.77,12.67,3.94,P<0.01).Compared with the model group,the levels of oxidation indices of MDA and ROS in the Bre intervention group decreased;the activity of antioxidant indices of SOD,and CAT,GSH levels,and GSH-related enzyme of GPx,GSR and GST activity increased.The effects of 25 and 50 mg/(kg·d)Bregroups were moreobvious(F=4.72,6.59,7.77,12.67,3.94,3.18,4.86,4.48,P<0.01).Conclusion Breviscapine can improve the injury of cardiomyocytes in rats with lead poisoning by inhibiting oxidative stress.
