槟榔苗对APV1病毒的生理响应

摘要 本研究以‘热研一号’槟榔苗作为实验材料,探究在海南大肆传播的由槟榔隐症病毒1(areca palm velarivirus 1,APV1)引起的槟榔黄化病致病机理。通过接种带APV1病毒粉蚧的方式获得携带病毒的槟榔苗,同时以健康槟榔苗(CK)作对照,在其他条件不变的基础上进行各项生理指标的测定和叶片显微结构的观察。研究结果表明,APV1病毒的侵染会导致槟榔苗叶肉变黄但是叶脉保持绿色,叶肉细胞密度、叶绿素a、叶绿素b和超氧化物歧化酶(SOD)活性均显著下降,叶绿体数量、过氧化物酶(POD)活性、过氧化氢酶(CAT)活性、脯氨酸含量、丙二醛含量则显著升高,叶片表皮厚度无显著变化。同时实验分高中低(L1,L2,L3)三个叶位进行对比测定。本研究对于以后防治槟榔黄化病具有一定的价值和意义。 In this study,'Reyan No.1'areca nut seedlings were used as experimental materials,to explore the pathogenic mechanism of areca nut chlorosis caused by Areca palm verarivirus 1(APV1)virus,which spread widely in Hainan.Betel nut seedlings infected with APV1 virus were obtained by inoculating mealybug.Meanwhile,healthy betel nut seedlings(CK)were used as control.On the basis of other conditions unchanged,the physiological indexes were determined and the microstructure of leaves was observed.The results showed that the infection of APV1 virus would cause the mesophyll of Areca catechu seedlings to turn yellow but the veins to remain green,and the mesophyll cell density,chlorophyll a,chlorophyll b and superoxide dismutase(SOD)activities decreased significantly,while chloroplast number,peroxidase(POD)activity,catalase(CAT)activity,proline content and malondialdehyde content increased significantly.There was no significant change in leaf epidermis thickness.At the same time,the experiment was divided into three leaf positions(L1,L2 and L3)for comparative determination.This study has certain value and significance for the control of betel nut yellowing disease in the future.
