柳茶多糖对高热环境秀丽隐杆线虫运动的影响及其机制研究

摘要 目的研究纯化后柳茶多糖对高热环境秀丽隐杆线虫运动的影响。方法超声辅助提取制备的柳茶多糖经DEAE Sepharose Fast Flow葡聚糖凝胶和Sephadex G100葡聚糖纯化后得到两种柳茶多糖(SLP-1和SLP-2)对其进行结构分析及高热线虫运动试验。结果SLP-1和SLP-2是13.5 kDa左右的含硫酸根的三螺旋结构酸性多糖。SLP-1多糖是由鼠李糖、葡萄糖、半乳糖、果糖和阿拉伯糖组成,而SLP-2多糖不含果糖。在提高秀丽隐杆线虫热刺激运动能力上,SLP-2表现更为优异,其机制是通过上调热休克因子1信使核糖核酸(heat shock factor 1 messenger ribonucleic acid,hsf-1 mRNA)来抵御高热环境对机体造成的损伤,上调热休克蛋白16.2信使核糖核酸(heat shock protein 16.2 messenger ribonucleic acid,hsp-16.2 mRNA)、热休克蛋白70信使核糖核酸(heat shock protein 70 messenger ribonucleic acid,hsp-70 mRNA)和热休克蛋白90信使核糖核酸(heat shock protein 90 messenger ribonucleic acid,hsp-90 mRNA)表达以保护衰老相关蛋白结构的稳定,还可通过调节核因子红细胞系2相关因子2-血红素加氧酶-1(nuclear factor erythroid 2-related factor 2-heme oxygenase 1,Nrf2-HO-1)途径以提高超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)的表达,从而提高抗氧化系统的总抗氧化能力(total antioxidant capacity,T-AOC)活性,,避免生成丙二醛(malondialdehyde,MDA)。结论柳茶多糖SLP-2是一种提高机体在高热环境中运动能力的功能性食品添加剂。 Objective To study the effects of purified polysaccharides from Sibiraea laevigata on the movement of Caenorhabditis elegans in high heat environment.Methods Two kinds of polysaccharides(SLP-1 and SLP-2)from Sibiraea laevigata,which were purified by DEAE Sepharose Fast Flow Glucan gel and Sephadex G100 Glucan after ultrasonic assisted extraction,they were analyzed in structure and tested on the movement of Caenorhabditis elegans in hot wire worms.Results SLP-1 and SLP-2 were about 13.5 kDa triple helix acidic polysaccharides containing rich sulfate.SLP-1 polysaccharide was composed of rhamnose,glucose,galactose,fructose and arabinose,while SLP-2 polysaccharide does not contain fructose.Compared with SLP-1,SLP-2 performs was better in improving the thermal stimulation of Caenorhabditis elegans.The mechanism was to up-regulate the expression of heat shock factor 1 messenger ribonucleic acid(hsf-1 mRNA)to resist the damage caused by high heat environment,and up-regulate the expression of heat shock protein 16.2 messenger ribonucleic acid(hsp-16.2 mRNA),heat shock protein 70 messenger ribonucleic acid(hsp-70 mRNA)and heat shock protein 90 messenger ribonucleic acid(hsp-90 mRNA)to protect the stability of aging related protein structure and regulate the nuclear factor erythroid 2-related factor 2-heme oxygenase 1(Nrf2-HO-1)pathway ability to promote the activity of superoxide dismutase(SOD)and catalase(CAT)antioxidant enzymes to improve the total antioxidant capacity(T-AOC)activity of antioxidant system to avoid the generation of MDA,so as to improve the exercise ability in high heat environment.Conclusion SLP-2 can be a functional food additive that can improve the athletic ability in high heat environment.
