摘要
为了探究RNA甲基转移酶NSun2在胃癌发生发展中的生物学功能和作用机制,尝试利用构建的过表达和敲低NSun2的胃癌细胞系,运用蛋白质双向电泳联合质谱的方法,寻找其潜在的下游底物.实验结果显示,在体外NSun2可以促进胃癌细胞的增殖,蛋白质组学方法筛选并鉴定出34个差异蛋白,其中蛋白谷胱甘肽S转移酶P(GSTP1-1)和蛋白激酶C抑制蛋白1(KCIP-1)可能是NSun2在胃癌中的潜在作用靶点.这对于认识胃癌发生发展的分子机制具有重要意义.
To explore the role and function of RNA methyltransferase NSun2 in gastric cancer,NSun2 overexpression and knockdown gastric cancer cell lines were constructed,and a combination of protein two-dimensional electrophoresis and mass spectrometry analysis was done to identify potential NSun2 targets.NSun2 was found to promote the proliferation of gastric cancer cells in vitro.A total of 34 proteins were identified after proteomics analysis,including glutathione S-transferase P(GSTP1-1)and kinase C inhibitor protein 1(KCIP-1),potential downstream targets of NSun2 in gastric cancer.Important insights into the molecular mechanisms underlying gastric cancer tumorigenesis and progression were gained.
