抗鼠棒状杆菌单克隆抗体制备及鉴定

摘要 以BALB/c小鼠为实验动物,将致敏的脾细胞与SP2/0瘤细胞进行融合,并采用间接ELISA进行克隆筛选,获得能分泌抗鼠棒状杆菌的特异性抗体且稳定生长的瘤细胞系。以此为基础,采用体内诱生腹水法制备单克隆抗体,测定其血清学效价、抗原反应灵敏度及亲和力。结果显示:制备的单抗血清学效价在1.6×10^(3)~1.28×10^(4),抗体水平均值非常高,灵敏度高,与类似抗原交叉反应率极低,具备构建快速检测法的应用前景。 BALB/c mice were used as experimental animals,sensitized spleen cells were fused with SP2/0 tumor cells,and indirect ELISA was used to clone and screen them,a stable tumor cell line that can secrete specific antibodies against Corynebacterium kutscheri was obtained.On the basis,the method of inducing ascites in vivo was used to prepare monoclonal antibodies,and the serological titer,antigen reaction sensitivity and affinity of the monoclonal antibodies were measured.The results showed that the serological titer of the prepared monoclonal antibodies was between 1.6×10^(3) and 1.28×10^(4),the average antibody level was very high,the sensitivity was high,and the cross reaction rate with similar antigens was very low,it has the application prospect of constructing a rapid detection method.
