摘要
目的探讨着丝粒蛋白K(CENPK)、干扰素诱导核蛋白16(IFI16)表达与中晚期宫颈癌患者预后的关系。方法选取行同步放化疗的106例中晚期宫颈癌患者宫颈癌组织,以同期40例因子宫脱垂或子宫肌瘤行全子宫切除的正常宫颈组织为对照。免疫组织化学检测组织中CENPK、IFI16蛋白表达。比较不同临床病理特征中晚期宫颈癌患者CENPK、IFI16蛋白表达差异。Kaplan-Meier方法分析CENPK、IFI16表达与中晚期宫颈癌预后的关系。Cox回归分析影响中晚期宫颈癌患者预后的影响因素。结果宫颈癌组织中CENPK、IFI16蛋白阳性表达率分别为70.75%(75/106)、67.92%(72/106),高于正常宫颈组织的10.00%(4/40)、15.00%(6/40),差异有统计学意义(P均<0.05)。不同肿瘤最大径、FIGO分期及淋巴结转移中晚期宫颈癌患者癌组织中CENPK、IFI16表达比较差异有统计学意义(P均<0.05)。CENPK阳性者5年累积生存率低于CENPK阴性者(P<0.05),IFI16阳性者5年累积生存率亦低于CENPK阴性者(P<0.05)。FIGO分期Ⅲ~Ⅳ、淋巴结转移、CENPK阳性表达、IFI16阳性表达是影响中晚期宫颈癌患者预后的独立因素(P均<0.05)。结论中晚期宫颈癌组织中CENPK、IFI16表达升高,二者表达与肿瘤最大径、FIGO分期及淋巴结转移有关,是影响患者不良预后的独立因素。
Objective To investigate the relationship between the expression of centromere protein K(CENPK)and interferon-inducible nuclear protein 16(IFI16)and prognosis of patients with advanced cervical cancer.Methods Totally 106 cases of cervical cancer tissues from patients with advanced cervical cancer who underwent concurrent radiotherapy and chemotherapy were selected as the study subjects,while 40 normal cervical tissues in patients undergoing total hysterectomy due to uterine prolapse or uterine fibroids during the same period were used as controls.CENPK and IFI16 protein expression levels in tissues were detected by immunohistochemistry.We compared the difference in the expression of CENPK and IFI16 protein in advanced cervical cancer patients with different clinicopathological features.Kaplan-Meier method was used to analyze the relationship between the expression of CENPK,IFI16 proteins and the prognosis of patients.Cox regression analysis was used to analyze the prognostic factors affecting patients with advanced cervical cancer.Results The positive rates of CENPK and IFI16 proteins in the cancer tissues were 70.75%(75/106)and 67.92%(72/106),respectively,which were higher than those[10.00%(4/40)and 15.00%(6/40)]of normal cervical tissues,with statistically significant differences(all P<0.05).The expression of CENPK and IFI16 in advanced cervical cancer tissues of patients with different maximum tumor sizes,FIGO staging,and lymph node metastasis showed statistically significant differences(all P<0.05).The 5-year cumulative survival rate of the CENPK positive group was significantly lower than that of the CENPK negative group,and the difference was statistically significant(P<0.05).The 5-year cumulative survival rate of the IFI16 positive group was significantly lower than that of the CENPK negative group,and the difference was statistically significant(P<0.05).FIGO stagingⅢ–Ⅳ,lymph node metastasis,positive expression of CENPK in cancer tissues,and positive expression of IFI16 were independent factors for the prognosis of patients with advanced cervical cancer(all P<0.05).Conclusion The expression of CENPK and IFI16 increases in middle to late stage cervical cancer,and their expression levels are related to maximum tumor size,FIGO staging,and lymph node metastasis,which are independent factors affecting poor prognosis of patients.
