PM_(2.5)协同不同饮食对大鼠糖脂代谢的影响

摘要 旨在研究细颗粒物(fine particulate matter,PM_(2.5))与多种饮食联合作用对机体糖脂代谢的影响。使用正常饮食(ND)、高脂肪饮食(HFD)、高碳水化合物饮食(HCD)喂养大鼠2个月,经气管内灌注暴露PM_(2.5)(7 mg·kg-1),观察PM_(2.5)暴露是否会引发糖脂代谢异常、肝损伤从而促进2型糖尿病的发展。结果表明,短期的PM_(2.5)暴露虽然不足以引起大鼠的糖耐量异常和胰岛素抵抗,但对氨基糖和核苷酸糖代谢途径有重要影响。结合病理学观察,发现PM_(2.5)造成ND组和HFD组的大鼠肝脏组织出现脂质空泡,血脂指标总胆固醇(TC)和甘油三脂(TG)显著升高;HCD组的大鼠肝脏细胞坏死,C-反应蛋白(CRP)水平升高。本研究考虑了饮食因素,进一步补充了PM_(2.5)健康效应的实验证据。 The purpose of this study was to investigate the effects of fine particulate matter(PM_(2.5))combined with multi-diet on glucose and lipid metabolism.Rats were fed with normal diet(ND),high-fat diet(HFD)and high-carbohydrate diet(HCD)for 2 months,and then exposed to PM_(2.5)(7 mg·kg-1)through intratracheal instillation to observe whether PM_(2.5) exposure can cause abnormal glucose and lipid metabolism,liver damage and promote the development of type 2 diabetes.The results showed that although short-term PM_(2.5) exposure was not sufficient to cause abnormal glucose tolerance and insulin resistance in rats,it had important effects on amino and nucleotide glucose metabolic pathways.Combined with pathological observation,it was found that PM_(2.5) caused lipid vacuole in liver tissues of rats in ND and HFD groups,and the levels of total cholesterol(TC)and triglyceride(TG)were significantly increased.In HCD group,liver cells were necrotic and C-reactive protein(CRP)level increased.This study takes dietary factors into account and adds to the experimental evidence for the health effects of PM_(2.5).
