神经生长因子对脂多糖引起的奶山羊支持细胞毒性抵抗研究

Study on the Cytotoxic Resistance of Nerve Growth Factor to Sertoli Cells Induced by Lipopolysaccharide in Dairy Goats

摘要 为探究神经生长因子(Nerve growth factor,NGF)对奶山羊支持细胞(Sertoli cells,SCs)的保护作用及机制,提取原代奶山羊SCs后用NGF处理,CCK-8检测发现,100~400 ng/mL的NGF能够显著提高细胞活力(P<0.05);此外,1000 ng/mL脂多糖(Lipopolysaccharide,LPS)处理SCs 24 h后细胞活力达到半数抑制效果。乳酸脱氢酶检测结果显示,NGF能够抑制LPS对SCs的毒性,其中,200 ng/mL NGF与LPS共处理6、12、24 h后,细胞毒性显著低于对照组(P<0.05),且200 ng/mL NGF对LPS的细胞毒性抵抗效果最佳。Western blot结果表明,NGF能够抑制LPS激活的PI3K/AKT/NFκB信号通路。综合以上研究,NGF能够通过PI3K/AKT/NFκB信号通路抑制LPS对SCs的细胞毒性。 In order to explore the protective effect and mechanism of nerve growth factor(NGF)on Sertoli cells(SCs)of dairy goats,the SCs were extracted and treated with NGF.CCk-8 assay showed that 100~400 ng/mL NGF significantly increased cell viability(P<0.05).In addition,1000 ng/mL Lipopolysaccharide(LPS)treatment for 24 h had half inhibition effect on SCs cell viability.The results of lactate dehydrogenase assay showed that NGF could inhibit the cytotoxicity of LPS to SCs,and 200 ng/mL NGF treated with LPS for 6,12,24 h significantly reduced the cytotoxicity of SCs compared with the control group(P<0.05),and had the best cytotoxic resistance to LPS.Western blot results showed that NGF significantly inhibited LPS-activated PI3K/AKT/NFκB signaling pathway(P<0.05).In conclusion,NGF can inhibit LPS cytotoxicity to SCs through PI3K/AKT/NFκB signaling pathway.
