胃腺癌中miR-550a-3p和TDP1 mRNA的表达水平及临床意义

The expression levels and clinical significance of miR-550a-3p and TDP1 mRNA in gastric adenocarcinoma

目的:分析胃腺癌中微小RNA-550a-3p(miR-550a-3p)和酪氨酰-DNA磷酸二酯酶1(TDP1)mRNA的表达水平及临床意义。方法:选取2020年1月至2022年10月在该院行手术治疗的54例胃腺癌患者进行前瞻性研究,留取术后胃腺癌组织和距肿物边缘>5 cm的正常胃黏膜组织,应用实时荧光定量PCR法检测胃腺癌组织和正常胃黏膜组织中miR-550-5p和TDP1 mRNA水平,比较胃腺癌组织与正常胃黏膜组织、不同病理特征胃腺癌组织miR-550a-3p和TDP1 mRNA水平,分析miR-550a-3p和TDP1 mRNA水平与胃腺癌发病的相关性。结果:胃腺癌组织miR-550a-3p和TDP1 mRNA水平均高于正常胃黏膜组织,差异有统计学意义(P<0.05);不同性别、年龄、淋巴结转移、分化程度、脉管侵犯的胃腺癌组织miR-550a-3p和TDP1 mRNA水平比较,差异无统计学意义(P>0.05);肿瘤最大径≥5cm、浸润深度浆膜及以下胃腺癌组织的miR-550a-3p和TDP1 mRNA水平均高于肿瘤最大径<5 cm、浸润深度未及浆膜的胃腺癌组织,差异有统计学意义(P<0.05);Pearson相关性分析结果显示,miR-550a-3p和TDP1 mRNA水平与胃腺癌发病呈正相关(r>0,P<0.05)。结论:miR-550a-3p和TDP1 mRNA水平与胃腺癌的病灶大小和浸润程度明显相关,且与发病情况呈正相关。

Objective:To analyze the expression levels and clinical significance of microRNA-550a-3p(miR-550a-3p)and tyrosyl-DNA phosphodiesterase 1(TDP1)in gastric adenocarcinoma.Methods:A prospective study was conducted on 54 patients with gastric adenocarcinoma who underwent surgical treatment in this hospital from January 2020 to October 2022.The postoperative gastric adenocarcinoma tissues and normal gastric mucosa tissues>5 cm from the edge of the tumor were collected.The levels of miR-550-5p and TDP1 mRNA in the gastric adenocarcinoma tissues and the normal gastric mucosa tissues were detected by real-time fluorescence quantitative PCR.The levels of miR-550a-3p and TDP1 mRNA in the gastric adenocarcinoma tissues and the normal gastric mucosa tissues as well as the gastric adenocarcinoma tissues with different pathological characteristics were compared.The correlations among the miR-550a-3p and TDP1 mRNA levels and gastric adenocarcinoma were analyzed.Results:The levels of miR-550a-3p and TDP1 mRNA in the gastric adenocarcinoma tissue were significantly higher than those in the normal gastric mucosal tissue,and the differences were statistically significant(P<0.05).There were no statistically significant differences in the miR-550a-3p and TDP1 mRNA levels among the gastric adenocarcinoma tissues of the patients with different genders,ages,lymph node metastasis,degree of differentiation,and vascular involvement(P>0.05).There were statistically significant differences in the miR-550a-3p and TDP1 mRNA levels between the gastric adenocarcinoma tissues with a maximum tumor diameter of≥5 cm and a depth of invasion of the serosa or below(P<0.05).Pearson correlation analysis showed the miR-550a-3p and TDP1 mRNA levels were positively correlated with gastric adenocarcinoma(r>0,P<0.05).Conclusions:The levels of miR-550a-3p and TDP1 mRNA are positively correlated with gastric adenocarcinoma.
