摘要
目的探讨转化生长因子β(TGF-β)对气道上皮-成纤维细胞互作的转录水平影响。方法BMT组(肺上皮细胞Beas-2b与肺成纤维细胞MRC5共培养并添加TGF-β)与BMP组(Beas-2b与MRC5共培养并添加磷酸盐缓冲液)细胞共培养后,DNBSEQ平台进行转录组测序。结果共获得16470个基因,1215个基因在BMT组中显著上调,1419个基因显著下调。与组织重塑、趋化、花生四烯酸代谢、炎症等相关基因在BMT组中的水平显著高于BMP组。差异基因富集分析显示,细胞外基质组织、细胞迁移、胶原纤维组织、层粘连蛋白结合、泛素蛋白连接酶结合等功能、组分显著富集。结论TGF-β影响上皮与成纤维细胞的互作,促进上皮-成纤维细胞互作模式转向细胞外基质积累、组织重塑和炎症表型。
OBJECTIVE To investigate the effect of TGF-β(transforming growth factor beta)on the interaction between airway epithelial cells and fibroblasts.METHODS After BMT group(co-cultured of lung epithelial cells Beas-2b and lung fibroblast MRC5 and added with TGF-β)and BMP group(Beas-2b co-cultured with MRC5 and added phosphate buffer solution)cells were co-cultured,the transcriptome sequencing(bulk RNA sequencing)of the co-cultured cells was performed using the DNBSEQ platform.RESULTS A total of 16470 genes were obtained,1215 genes were significantly upregulated in the BMT group,and 1419 genes were significantly downregulated in the BMT group.The expression levels of genes related to tissue remodeling,cell chemotaxis,arachidonic acid metabolism and inflammation were significantly higher in the BMT group than in the BMP group.Enrichment analysis of differential genes showed that extracellular matrix organization,cell migration,collagen fibril organization,laminin binding and ubiquitin protein ligase binding were significant enrichment.CONCLUSION TGF-βaffects epithelial-fibroblast interactions,promoting epithelial-fibroblast interaction patterns toward extracellular matrix accumulation,tissue remodeling,and inflammatory phenotypes.
