转铁蛋白受体1和血管内皮生长因子在儿童及青少年骨肉瘤患者中的表达及临床意义

Expression and clinical significance of transferrin receptor 1 and vascular endothelial growth factor in children and adolescents with osteosarcoma

摘要 目的 探讨转铁蛋白受体1(TFR1)和血管内皮生长因子(VEGF)在儿童及青少年骨肉瘤患者中的表达及临床意义。方法 收集35例儿童及青少年骨肉瘤患者的骨肉瘤组织和癌旁正常组织,采用免疫组化染色法检测TFR1、VEGF表达情况。比较骨肉瘤组织和癌旁正常组织中TFR1和VEGF表达情况,比较不同临床特征儿童及青少年骨肉瘤患者骨肉瘤组织中TFR1和VEGF表达情况,比较不同TFR1、VEGF表达情况儿童及青少年骨肉瘤患者的生存情况,采用Spearman相关分析法分析TFR1与VEGF表达的相关性。结果 儿童及青少年骨肉瘤患者骨肉瘤组织中TFR1和VEGF中高表达率均明显高于癌旁正常组织,差异均有统计学意义(P﹤0.01)。中低分化、有远处转移、Enneking分期为Ⅱ~Ⅲ期儿童及青少年骨肉瘤患者骨肉瘤组织中TFR1、VEGF中高表达率均高于高分化、无远处转移、Enneking分期为Ⅰ期的患者,差异均有统计学意义(P﹤0.05)。TFR1、VEGF低表达儿童及青少年骨肉瘤患者的生存情况均优于TFR1、VEGF中高表达患者,差异均有统计学意义(P﹤0.05)。儿童及青少年骨肉瘤患者中TFR1与VEGF的表达呈正相关(r=0.669,P﹤0.05)。结论 TFR1和VEGF在儿童及青少年骨肉瘤患者中表达上调,可能参与骨肉瘤的发生、发展、侵袭和远处转移,可作为骨肉瘤诊断、恶性程度判断及预后评估指标。 Objective To investigate the expression and clinical significance of transferrin receptor 1(TFR1) and vascular endothelial growth factor(VEGF) in children and adolescents with osteosarcoma.Method The osteosarcoma tissues and adjacent normal tissues of 35 children and adolescents with osteosarcoma were collected.The expression of TFR1 and VEGF were detected by the immunohistochemical staining method.The expression of TFR1 and VEGF in osteosarcoma tissues and adjacent normal tissues were compared.The expression of TFR1 and VEGF in osteosarcoma tissues of children and adolescents with different clinical characteristics were compared.The survival of children and adolescents with osteosarcoma with different TFR1 and VEGF expression were also compared.The correlation between TFR1and VEGF expression was analyzed by Spearman correlation analysis.Result The medium and high expression rates of TFR1 and VEGF in osteosarcoma tissues of children and adolescents with osteosarcoma were significantly higher than those in adjacent normal tissues,and the differences were statistically significant(P<0.01).The medium and high expression rates of TFR1 and VEGF in osteosarcoma tissues from children and adolescents with poorly differentiated,distant metastasis,and Enneking stage Ⅱ-Ⅲ osteosarcoma were higher than those of well-differentiated,no distant metastasis,Enneking stage I patients,and the differences were statistically significant(P<0.05).The survival of children and adolescents with osteosarcoma with low expression of TFR1 and VEGF were better than those of patients with high expression of TFR1 and VEGF,and the differences were statistically significant(P<0.05).The expression of TFR1 and VEGF were positively correlated in children and adolescents with osteosarcoma(r=0.669,P<0.05).Conclusion TFR1 and VEGF are up-regulation in children and adolescents with osteosarcoma.The two indicators may be involved in the occurrence,development,invasion,and distant metastasis of osteosarcoma,and could be used to diagnose osteosarcoma,estimate malignant degree and prognosis of osteosarcoma.
