摘要
目的 基于中医古籍数据挖掘及网络药理学研究,探讨贯众治疗崩漏的价值及其潜在机制。方法 以《中华医典》为数据源检索贯众的功效及应用情况,采用Excel做数据处理,并导入古今医案云平台做统计分析。使用TCMSP检索贯众的成分及相应靶点,使用GeneCards检索异常子宫出血的疾病靶点,使用Metascape做GO和KEGG富集分析,并借助VENNY、Cytoscape、微生信网站做数据可视化分析。结果 中医古籍数据挖掘结果显示,贯众单味药处方176频次,主治病证38种,其中崩漏为贯众单味药内服应用频次最高的疾病;贯众组方991频次,主治病证127种,其中崩漏应用频次排序第8。贯众治疗崩漏配伍的核心药物为当归、芍药、白术、黄芪、人参、荆芥。网络药理学研究结果显示,贯众中山奈酚、白绵马素AA等有效成分可能通过F2、F7、F10、F11、PLG、ANXA5、CTSK、PLAT、CYP19A1、CYP1A1、CYP1B1、ESR1、ESR2、PGR、TNF等基因靶点调控补体和凝血级联反应、卵巢类固醇生成、TNF、HIF-1等信号通路发挥治疗异常子宫出血的作用。结论 贯众能凉血止血、收敛止血、散瘀止血,用治崩漏有效。贯众可通过多成分、多靶点、多通路发挥其治疗崩漏的作用。
Objective To explore the value and underlying mechanism of Guanzhong in the treatment of metrorrhagia and metrostaxis based on data mining from traditional Chinese medicine(TCM)ancient texts and network pharmacology.Methods The efficacy and applications of Guanzhong were retrieved from the Chinese Medical Classics.Excel was used for data processing,and the data were analyzed using the Ancient and Modern Medical Case Cloud Platform.TCMSP was used to retrieve the components and corresponding targets of Guanzhong,while GeneCards was used to retrieve disease targets related to metrorrhagia and metrostaxis.GO and KEGG enrichment analyses were performed using Metascape,and data visualization analysis was carried out using VENNY,Cytoscape,and the bioinformatics website.Results Data mining from TCM ancient texts revealed that the frequency of Guanzhong reached 176 times in single-herb prescriptions,targeting 38 different diseases,with metrorrhagia and metrostaxis being the most frequently mentioned disease.In compound formulas,Guanzhong appeared in 991 prescriptions,targeting 127 different diseases,with metrorrhagia and metrostaxis being the 8th most frequently treated disease.Core herbs frequently used in conjunction with Guanzhong for treating metrorrhagia and metrostaxis included Angelicae Sinensis Radix,Paeoniae Radix Alba,Atractylodis Macrocephalae Rhizoma,Astragali Radix,Ginseng Radix et Rhizoma,and Schizonepetae Herba.Results from network pharmacology suggested that effective compounds in Guanzhong such as kaempferol and albaspidin AA might regulate complement and coagulation cascades,ovarian steroidogenesis,TNF,HIF-1,and other signaling pathways through genes like F2,F7,F10,F11,PLG,ANXA5,CTSK,PLAT,CYP19A1,CYP1A1,CYP1B1,ESR1,ESR2,PGR,and TNF,thereby exerting the role in the treatment of metrorrhagia and metrostaxis.Conclusion Guanzhong possesses properties of cooling the blood,astringing,and dissipating stasis to stop bleeding,and is effective in treating metrorrhagia and metrostaxis.Guanzhong exerts its therapeutic effect on metrorrhagia and metrostaxis through multiple components,multiple targets,and multiple pathways.
