摘要
目的:探究微小miR-184(miR-184)在调控女性滋养层细胞凋亡中的作用及其分子机制。方法:利用定量的反转录(PCR)技术检测62例女性滋养层细胞组织中微小miR-184的表达,以分析和观察女性滋养层细胞组织中的miR-184在人绒毛膜滋养层细胞(HTR8/Svneo)和滋养层细胞系(Swan71)两种细胞中的表达水平和对细胞增殖与凋亡的作用机制。结果:miR-184在人绒毛膜滋养层细胞(HTR8/Svneo)和人滋养层细胞(Swan71)两种细胞中的相对表达水平在滋养层细胞中比在正常的上皮组织细胞中更高(均P<0.05);转染小干扰RNA(siRNA)沉默miR-184促使miR-184的表达能够抑制HTR8/SVneo和Swan71细胞的增殖,利用si-miR-184的转染降低miR-184的表达水平能够阻碍女性滋养层细胞的凋亡(P<0.01),而miR-184的过度表达能够明显推动女性滋养层细胞的凋亡(P<0.01)。结论:通过抑制miR-184表达可以调控和诱导滋养层细胞凋亡的保护作用。
Objective:To explore the role and molecular mechanism of microRNA-184(miR-184)in regulating apoptosis of female trophoblast cells.Methods:The expression of miR-184 in 62 female trophoblastic tissues was detected by quantitative reverse transcription-PCR in order to analyze and observe the expression level of miR-184 in HTR8/SVneo and Swan71 cells and the mechanism of its effect on cell proliferation and apoptosis.Results:The relative expression level of miR-184 in HTR8/SVneo and Swan71 cells was higher in trophoblastic cells than in normal epithelial cells(all P<0.05).Transfection of si-miR-184 can promote the expression of miR-184 and inhibit the proliferation of HTR8/SVneo and Swan71 cells.Using si-miR-184 to reduce the expression level of miR-184 can inhibit the apoptosis of female trophoblast cells(P<0.01),while the overexpression of miR-184 can significantly promote the apoptosis of female trophoblast cells(P<0.01).Conclusion:Inhibiting the expression of miR-184 can regulate and induce the protective effect of trophoblast cell apoptosis.
