茅岩莓提取物治疗放射性口腔溃疡的作用研究

摘要 目的采用X射线复制大鼠放射性口腔溃疡模型,对茅岩莓提取物治疗放射性口腔溃疡的效果进行评价。方法SD大鼠60只,雌雄各半,随机分为正常对照组,模型对照组,金因肽组(5000 IU·kg^(-1)),茅岩莓提取物低、中、高(16、32、64 mg·kg^(-1))剂量组,每组10只。各组大鼠按15 Gy剂量采用X射线照射大鼠口部及内侧口腔黏膜,复制放射性口腔溃疡模型。分别于造模后24 h进行药物干预,2次·d^(-1),连续14 d。每日对各组动物进行一般临床观察,定期监测体重、摄食量及口腔黏膜评分,末次给药后采集各组大鼠口腔黏膜,检测口腔黏膜组织中IL-6、TNF-α、IL-1β的含量,并进行组织病理学检查。结果金因肽和低、中、高剂量茅岩莓提取物均可提高动物体重增长率和摄食量;金因肽和中、高剂量茅岩莓提取物可显著降低口腔黏膜评分和IL-6表达(P<0.05,P<0.01),减轻口腔黏膜病变程度;金因肽和高剂量茅岩莓提取物可显著降低口腔黏膜组织IL-1β表达(P<0.05,P<0.01)。结论茅岩莓提取物对放射性口腔溃疡具有修复作用,其作用可能与调节口腔黏膜炎症因子的水平相关。 Objective To replicate the rat radioactive oral ulcer model with X-rays and evaluate the effect of thatch berry extract on radioactive oral ulcer.Methods Totally 60 SD rats,30 males and 30 females,were randomly divided into a normal control group,a model control group,a Gene Time group,and low(16 mg·kg^(-1)),medium(32 mg·kg^(-1))and high(64 mg·kg^(-1))dose groups of thatch berry extract(10 rats in each group).Rats in each group were irradiated with X-rays at a dose of 15 Gy in the mouth and medial oral mucosa to replicate the radioactive oral ulcer model.Pharmacological intervention was administered 24 h after the modelling,twice daily for 14 days.The rats in each group were observed daily,and their body weight,food intake and oral mucosa scores were monitored regularly.After the last dose,the rats in each group were euthanized by bleeding from the abdominal aorta,and the levels of IL-6,TNF-αand IL-1βin the oral mucosa tissues were measured and histopathologically examined.Results Gene Time and all doses of thatch berry extract increased body weight growth rate and food intake.Gene Time,medium and high doses of thatch berry extract significantly reduced the oral mucosal score and IL-6 expression(P<0.05,P<0.01),and reduced the degree of oral mucosal lesions.Gene Time and high doses of thatch berry extract significantly reduced oral mucosal score and IL-1βexpression(P<0.05,P<0.01).Conclusion The repairing effect of thatch berry extract might be related to the modulation ofllevels of inflammatory factors in the oral mucosa.
