一测多评法同时测定羌活胜湿汤中6种成分的含量

摘要 目的建立一测多评法(QAMS)同时测定羌活胜湿汤中绿原酸、升麻素苷、阿魏酸、洋川芎内酯Ⅰ、5-O-甲基维斯阿米醇苷、甘草酸铵的含量。方法采用COSMOSIL Packed Column 5C18-MS-Ⅱ色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.2%甲酸水溶液为流动相,梯度洗脱,流速1.0 mL·min^(-1),柱温30℃,检测波长254 nm。结果各成分在各自质量浓度范围内与峰面积线性关系良好(R2>0.999)。以甘草酸铵为参照物,绿原酸、升麻素苷、阿魏酸、洋川芎内酯Ⅰ、5-O-甲基维斯阿米醇苷的校正因子分别为0.6155、0.1310、0.4729、0.2063、0.0969,并计算各成分含量,同时采用外标法(ESM)测定6个成分的含量,两种方法测定的结果差异无统计学意义。结论建立了羌活胜湿汤中绿原酸、升麻素苷、阿魏酸、洋川芎内酯Ⅰ、5-O-甲基维斯阿米醇苷、甘草酸铵6个成分的QAMS测定方法,该法可用于羌活胜湿汤的质量控制。 Objective To establish a quantitative analysis of multi-components by single markers(QAMS)to determine the content of chlorogenic acid,coenoside,ferulic acid,ligustratus I,5-O-methylvesamitol side and ammonium glycyrrhizate in Qianghuo Shengshi decoction.Methods The determination was performed on COSMOSIL Packed Column 5C18-MS-Ⅱ(250 mm×4.6 mm,5μm)with acetonitrile-0.2%formic acid as the mobile phase in gradient elution.The volume flow rate was 1.0 mL·min^(-1).The column temperature was 30℃,and the detection wavelength was 254 nm.Results In the linear range,all components had good linearity(R2>0.999).With ammonium glycyrrhizate as the internal standard,the correction factors of chlorogenic acid,cohorin,ferulic acid,ligustrine I and 5-O-methylvesamitol side were 0.6155,0.1310,0.4729,0.2063 and 0.0969,respectively.The content of the 6 components was also determined by the external standard method(ESM).The results of QAMS and ESM had no significant difference.Conclusion The QAMS method of chlorogenic acid,coenoside,ferulic acid,ligustratus I,5-O-methylvesamitol side and ammonium glycyrrhizate in Qianghuo Shengshi decoction has been established.QAMS can be used for the quality control of Qianghuo Shengshi decoction.
