基于非靶向代谢组学分析不同陈化时间老鹰茶代谢产物的差异

Difference analysis of metabolites in hawk tea with different aging time based on non-targeted metabolomics technology

摘要 为探究陈化时间对老鹰茶代谢物的影响,基于超高效液相色谱-四极杆-飞行时间质谱(UPLC-QTOF-MS)技术结合主成分分析(PCA)及正交偏最小二乘-判别分析(OPLS-DA)法对未陈化、陈化1年、2年和3年的老鹰茶代谢物进行分析,并以变量投影重要度(VIP)≥1且P<0.05筛选差异显著代谢物。结果表明,不同老鹰茶样本中共注释到4530个代谢物;与未陈化样本相比,经过陈化后老鹰茶样本中代谢物种类及含量有明显变化,但不同陈化时间样本代谢物种类和含量差异较小;经过1、2、3年陈化的老鹰茶样本的差异显著代谢物数分别为832个、925个和1071个。陈化后老鹰茶样本中与鲜味相关的L-天冬氨酸含量增加,与苦味相关的次黄嘌呤和7-甲基黄嘌呤含量降低,野漆树苷等黄酮苷类化合物的含量逐渐增加。该研究表明,随陈化时间的增加,一定程度上提升了老鹰茶的滋味与营养价值。 In order to explore the effects of aging time on metabolites of Hawk tea,the metabolites of unaged,aged 1 year,2 years and 3 years of Hawk tea were analyzed based on ultra-high performance liquid chromatography-Quadrupole-time-of-flight mass spectrometry(UPLC-QTOF-MS)combined with principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA),the metabolites with significant differences were screened with variable importance in the projection(VIP)≥1 and P<0.05.The results showed that a total of 4530 metabolites were annotated in different Hawk tea samples.Compared with the samples without aging,there were obvious changes in the metabolite types and contents in the Hawk tea samples after aging,but there was little difference in the metabolite types and contents in the samples with different aging times.The metabolites with significant difference in the tea Hawk samples after aging for 1,2 and 3 years were 832,925 and 1071,respectively.After aging,the contents of L-aspartic acid related to umami increased,the contents of hypoxanthine and 7-methylxanthine related to bitter taste decreased,and the contents of flavonoid glycosides such as rhoifolin gradually increased.The study implies that with the increase of aging time,the taste and nutritional value of eagle tea were improved to a certain extent.
