miR-218-5p/GTSE1调控轴抑制肺腺癌细胞的增殖、迁移及侵袭

MiR-218-5p/GTSE1 axis suppresses proliferation,invasion and migration of lung adenocarcinoma cells

目的探讨GTSE1在肺腺癌中的分子机制,以及其在细胞增殖、迁移和侵袭中的作用。方法将PC-9和A549细胞分为多组,进行过表达或沉默GTSE1、miR-218-5p。通过TCGA数据确定和评估miR-218-5p和靶基因GTSE1在肺腺癌中的预后意义。qRT-PCR和Western blot分别检测GTSE1和miR-218-5p的mRNA和蛋白表达水平。CCK-8、克隆形成、Transwell实验检测细胞增殖、迁移和侵袭能力。双荧光素酶报告实验分析miR-218-5p和GTSE1的靶向关系。结果GTSE1在肺腺癌组织和细胞中高表达,且高表达GTSE1的肺腺癌患者预后不良。过表达GTSE1可促进肺腺癌细胞的增殖、迁移与侵袭。另外,miR-218-5p在肺腺癌组织和细胞中低表达,并且能够靶向下调GTSE1的表达。实验结果显示,高表达miR-218-5p能够抑制肺腺癌细胞的增殖、迁移与侵袭,而其抑制作用能够被GTSE1过表达处理所恢复。结论miR-218-5p/GTSE1轴能够抑制肺腺癌细胞的增殖、迁移与侵袭。

Objective To investigate the molecular mechanism of G 2 and S phase-expressed-1(GTSE1)in lung adenocarcinoma(LUAD)and its role in cell proliferation,migration and invasion.Methods PC-9 and A549 cells were divided into multiple groups to overexpress or silence GTSE1 and miR-218-5p.The prognostic significance of miR-218-5p and target gene GTSE1 in lung adenocarcinoma was determined and evaluated using TCGA data.The mRNA and protein expression levels of GTSE1 and miR-218-5p were detected by qRT-PCR and Western blot,respectively.CCK-8,clonal formation and Transwell assay were used to detect cell proliferation,migration and invasion.Dual luciferase assay to analyze the targeting relationship between miR-218-5p and GTSE1.Results GTSE1 was highly expressed in LUAD tissues and cells,and the prognosis of LUAD patients with high GTSE1 expression was poor.Overexpression of GTSE1 could facilitate proliferation,invasion and migration of LUAD cells.In addition,miR-218-5p was lowly expressed in LUAD tissues and cells,and could target and down-regulate GTSE1 expression.The results of the rescue experiment showed that high expression of miR-218-5p could inhibit the proliferation,invasion and migration of LUAD cells,and its inhibitory effect could be restored by GTSE1 overexpression treatment.Conclusion MiR-218-5p/GTSE1 could suppress proliferation,invasion and migration of LUAD cells.
