糖通饮对糖尿病肾病大鼠肾脏纤维化的延缓作用 被引量:2

摘要 目的探讨糖通饮延缓糖尿病肾病大鼠肾脏纤维化的作用机制。方法随机抽取10只大鼠为正常组,其余50只采用高脂高糖饮食联合STZ多次注射进行糖尿病肾病造模,造模成功后随机分为模型组,厄贝沙坦组,糖通饮低、中、高剂量组,每组10只。各组大鼠给予对应药物干预8周后,代谢笼收集大鼠24 h尿液,股动脉取血,处死大鼠并取双侧肾组织。检测各组大鼠肾脏肥大指数、空腹血糖、24 h尿微量白蛋白、血尿素氮、血肌酐;HE、Masson染色观察各组大鼠肾组织形态学变化;RT-qPCR法检测miRNA-21、TGF-β_(1)、Col-Ⅲ、FN mRNA表达;Western blot法及免疫组化染色法检测肾组织TGF-β_(1)、Col-Ⅲ、FN蛋白表达。结果与模型组比较,糖通饮各剂量组大鼠肾脏肥大指数、空腹血糖、24 h尿微量白蛋白、血尿素氮、血肌酐水平降低(P<0.05),糖通饮中、高剂量组改善效果优于糖通饮低剂量组(P<0.05);光镜下厄贝沙坦组与糖通饮各剂量组大鼠肾组织病理形态得到不同程度改善;糖通饮中、高剂量组与厄贝沙坦组miRNA-21表达及TGF-β_(1)、Col-Ⅲ、FN mRNA和蛋白表达降低(P<0.05)。结论糖通饮能够延缓糖尿病肾病大鼠肾脏纤维化,其机制可能是通过对miRNA-21、TGF-β_(1)调控产生的。 AIM To explore the effect and mechanism of Tangtongyin on delaying renal fibrosis in diabetic kidney disease rats.METHODS Ten rats were randomly selected as the normal group,and the other 50 rats were treated with high-fat and high-sugar diet combined with STZ multiple low-dose injections to establish diabetic kidney disease model.After successful modeling,the rats were randomly divided into model group,irbesartan group,low-dose,middle-dose group and high-dose group of Tangtongyin with 10 rats in each group.After 8 weeks of corresponding intervention in each group,the rats in each group were sacrificed and bilateral renal tissues were taken after 24 h urine was collected by metabolic cage and blood was taken from femoral artery.And the renal hypertrophy index,FBG,24 h ALB,BUN and SCr of rats in each group were detected.HE and Masson staining were used to observe the morphological changes of kidney in each group.The mRNA expressions of miRNA-21,TGF-β_(1),Col-Ⅲand FN were detected by RT-qPCR.The protein expressions of TGF-β_(1),Col-Ⅲand FN in renal tissues were detected by Western blot and immunohistochemical staining.RESULTS Compared with the model group,the renal hypertrophy index,FBG,24 h ALB,BUN and SCr of rats were all decreased in each dose group of Tangtongyin(P<0.05).The improvement effect of middle and high dose groups of Tangtongyin were better than that of low dose group(P<0.05).Compared with the model group,the pathological morphology of kidney tissue in Tangtongyin and Irbesartan groups was improved in different degrees under light microscope.The expression of miRNA-21 and the mRNA and protein expressions of TGF-β_(1),Col-Ⅲand FN in kidney tissue of the middle and high dose groups of Tangtongyin and Irbesartan groups were decreased(P<0.05).CONCLUSION Tangtongyin can delay renal fibrosis in diabetic kidney disease rats,and its mechanism may be related to the regulation of miRNA-21 and TGF-β_(1).
