高脂饮食诱导的肥胖小鼠肝脏转录组分析

摘要 目的对长期高脂饮食饲喂构建的肥胖小鼠肝脏组织进行转录组分析,探讨肝脏功能对肥胖发生的作用机制。方法利用长期高脂饮食饲喂构建小鼠肥胖模型,通过形态观察,体重、体脂及血糖检测进行模型评价。建模成功后对小鼠脂肪组织和肝脏组织进行HE染色,观察组织病理改变,并通过转录组检测小鼠肝脏组织基因表达变化。结果高脂饮食饲喂小鼠8周后小鼠体重及血糖水平明显增加,脂肪组织脂质累积,脂肪细胞肥大,呈现肥胖表型。对肝脏组织进行转录组测序分析发现肥胖小鼠肝脏组织基因表达存在明显改变,基于P_adjust≤0.05和log2FC值筛选获得一些关键差异表达基因供后续深入研究。结论肝脏功能在肥胖及其相关疾病发生发展中发挥重要作用。 Objective To explore the mechanism of liver function in obesity through liver tissue transcriptome analysis in obese mice with long-term high-fat diet feeding.Methods In order to establish a mouse obesity model,mice were subjected to long-term high-fat diet.The model was evaluated by morphological observation,measurement of body weight and body fat,and blood glucose levels.After successful modeling,the pathological changes of adipose tissue and liver tissue were observed by HE staining,and the changes of gene expression in liver tissue of mice were detected by transcriptome analysis.Results After 8 weeks of high-fat diet feeding,mice exhibited significant increases in body weight and blood glucose levels.Moreover,there was noticeable lipid accumulation and hypertrophy of adipose cells in mouse adipose tissue,leading to the development of obesity.Transcriptome sequencing analysis of liver tissue revealed significant changes in functional gene expression of obese mouse,and several key differentially expressed genes were screened based on P_adjust≤0.05 and log2FC value for further study.Conclusion Liver function may play an important role in the development of obesity.
