两种玉竹根茎显微形态及同工酶谱带比对研究

摘要 本文利用石蜡切片法和同工酶谱带技术对2种玉竹形态学和显微结构、同工酶谱带的差异对2种玉竹遗传特性进行鉴定。研究发现,2种玉竹石蜡切片显微结构差异显著,北方产区玉竹的表皮厚度平均为2.24×10^(-2)mm;薄壁组织厚度平均为8.69×10^(-2)mm;微管组织厚度平均为1.561 mm;南方产区玉竹的表皮厚度平均为3.85×10^(-2)mm;薄壁组织厚度平均为5.36×10^(-2)mm;微管组织厚度平均为3.114mm;POD同工酶谱带相似,共分离出4条谱带,且所有谱带的Rf值都小于0.5,这可能由于宽甸玉竹中的某些蛋白含量低于邵县玉竹的原因。结果表明,2种玉竹存在种内遗传多样性差异,本研究对北方产地玉竹与南方产地玉竹根茎显微结构及同工酶谱带进行比对分析,为我国玉竹资源进一步开发利用奠定理论基础。 In this paper,the differences of morphology,microstructure and isozyme bands of Polygonatum were analyzed by using the paraffin section method and isozyme band technique,and the genetic characteristics of two Polygonatum were identified.The results showed that the micro structures of the paraffin sections of the two species were significantly different,and the epidermal thickness of two species was 2.24×10^(-2)mm on average.The average thickness of parenchyma was 8.69×10^(-2)mm.The average thickness of microtubule tissue was1.561 mm.The average thickness of epidermis was 3.85×10^(-2)mm in south China.The average thickness of parenchyma was 5.36×10^(-2)mm.The average thickness of microtubule tissue was 3.114 mm.POD isozyme bands were similar,and a total of 4 bands were isolated,and Rf values of all bands were less than 0.5,which might be due to some protein content in Kuandian Polygonatum was lower than ShaoyangPolygonatum.The results showed that there were differences in intraspecies genetic diversity between the two species.In this paper,the microstructure and isozyme bands of the rhizome of the two species were analyzed for the first time,which would lay a theoretical foundation for the further development and utilization of the resources of the two species.
