摘要
利用PacBio三代测序平台测定散纹盛蛱蝶成虫肠道细菌的16S rRNA基因全序列,应用SMRTLINK(v9)和USEARCH(v10)软件获得一致性CCS序列和操作分类单元数量,初步分析其肠道细菌群落结构和Alpha多样性。结果表明,散纹盛蛱蝶的3个样本共获得48458条CCS序列,总长度69573368 bp,经优化后获得35586条有效reads,在98.65%的相似度下被聚类为2547个OTUs,共注释到9个门,15个纲,25个目,48个科,71个属,217个种。在门分类阶元上,以变形菌门为优势门,平均相对丰度为55.89%;在属分类阶元上,以沉积物杆状菌属为优势属,平均相对丰度为21.24%。Alpha多样性分析结果显示,散纹盛蛱蝶成虫肠道细菌Chao指数、Ace指数、Shannon指数和Simpson指数分别为1737.1410~2095.6292、1943.0551~2321.3013、6.7782~7.3076和0.0246~0.0540,表明散纹盛蛱蝶成虫肠道细菌群落有较高的丰富度和多样性,为进一步研究昆虫肠道细菌的功能及其在生物防治中的应用提供理论参考依据。
The complete sequences of the 16S rRNA genes of intestinal bacteria in the adult of Symbrenthia lilaea were determined by PacBio RSⅡ.The numbers of circular consensus sequence(CCS)and operational taxonomic units(OTUs),species composition,abundance and diversity of the bacteria were analyzed by using SMRTLINK(v9)and USEARCH(V10)softwares.A total of 48,458 CCS sequences with a total length of 69,573,368 bp were obtained in the three samples of S.lilaea.After filtration,a total of 35,586 reads were obtained and clustered into 2,547 OTUs with 98.65% similarity.These OTUs were annotated into 9 phyla,15 classes,25 orders,48 families,71 genera and 217 species.The Proteobacteria was the most dominant bacteria with an average relative abundance of 55.89%.At the genus level,Sediminibacterium was the most dominant bacteria(21.24%).The indices of Chao,Ace,Shannon and Simpson were 1737.1410~2095.6292,1943.0551~2321.3013,6.7782~7.5921 and 0.0246~0.0540,respectively.The results of Alpha diversity indices showed that the bacteria in the intestine of adult S.lilaea are diverse.The information of bacterial abundance and diversity in the intestine of S.lilaea will facilitate the studies on the new technology of its control based on the intestinal bacteria.
