加味理中汤方对脾虚泄泻小鼠血常规、血糖及脏器指数的影响

Effect of Jiawei Lizhong Decoction on blood routine,blood glucose and organ index in mice with diarrhea due to spleen deficiency

试验旨在研究加味理中汤方对脾虚泄泻模型小鼠血常规、血糖及脏器指数的影响。试验中复方由干姜、党参、白术、甘草、茯苓、山药、木香、桂枝、砂仁组成。选择无特定病原体级ICR小鼠72只,随机分为6组,每组12只小鼠,雌、雄各半。除空白对照组(C组)小鼠灌胃同体积的生理盐水外,其余各组小鼠灌胃番泻叶提取物9 d建立脾虚泄泻模型。试验第10 d停用番泻叶,恩诺沙星治疗组(A组)小鼠灌胃恩诺沙星水溶液,各中药组分别给予低剂量(6.5 g/kg)、中剂量(13.0 g/kg)、高剂量(26.0 g/kg)灌胃治疗,C组和泄泻模型组(M组)用同体积的生理盐水灌胃,连续治疗3 d。结果显示,与C组相比,M组小鼠空腹血糖含量、淋巴细胞百分比、肝脏指数均极显著降低(P<0.01),血小板数目显著降低(P<0.05),中性粒细胞百分比及白细胞数目显著增加(P<0.01)。与M组比,各治疗组小鼠中性粒细胞百分比极显著降低(P<0.01),淋巴细胞百分及空腹血糖含量极显著升高(P<0.01);中药高剂量组(ZH组)小鼠血小板数目显著提高(P<0.05),肝脏指数极显著提高(P<0.01)。与A组比,ZH组小鼠血糖含量、肝脏指数均极显著升高(P<0.01),血小板数目、肾脏指数均显著升高(P<0.05)。研究表明,加味理中汤方对脾虚泄泻小鼠的血常规、血糖及肝肾脏器指数等指标异常具有一定的改善作用,高剂量中药效果最佳且优于恩诺沙星。

The aim of the experiment was to study the effect of Jiawei Lizhong Decoction on blood routine,blood glucose and organ index in mice model of splenic deficiency diarrhea.The compound was composed of Rhizoma zingiberis,Codonopsis pilosula,Atractylodes macrocephala,licorice,tuckahoe,yam,Aucklandiae Radix,cassia twig and Fructus amomi.A total of 72 mice without specific pathogen grade ICR were randomly divided into six groups with 12 mice in each group(half female and half male).Except for the mice in blank control group(group C),the mice in the other groups were administrated with Folium sennae for 9 d to establish spleen deficiency diarrhea model.On the 10th day of the experiment,Folium sennae was stopped,enrofloxacin treatment group(group A)was given enrofloxacin aqueous solution,and the Chinese medicine groups were given low dose(6.5 g/kg),medium dose(13.0 g/kg)and high dose(26.0 g/kg),respectively,group C and diarrhea model group(group M)were given the same volume of normal saline.The treatment was continued for 3 d.The results showed that compared with group C,fasting blood glucose content,lymphocyte percentage and liver index in group M were extremely decreased(P<0.01),platelet number was significantly decreased(P<0.05),neutrophil percentage and white blood cell number were extremely increased(P<0.01).Compared with group M,the percentage of neutrophil in each treatment group was extremely decreased(P<0.01),and the percentage of lymphocyte and fasting blood glucose were extremely increased(P<0.01).The number of platelet was significantly increased in the high dose group(ZH group)(P<0.05),and the liver index was extremely increased(P<0.01).Compared with group A,the blood glucose content and liver index of mice in ZH group were extremely increased(P<0.01),and the platelet number and kidney index were significantly increased(P<0.05).The study indicates that Jiawei Lizhong Decoction can ameliorate the abnormal blood routine,blood glucose and liver and kidney indexes in mice with splenic deficiency and diarrhea,and the effect of high dose Chinese medicine was the best and better than enrofloxacin.
