温度和酸性环境对A型塞内卡病毒稳定性的影响

摘要 A型塞内卡病毒(SVA)作为猪水疱性传染病的病原体之一,日益影响我国生猪养殖业的发展。为探究温度和酸性环境对SVA复制的影响,以CHN/SVA/HB/2018 SVA株作为研究材料,采用生物信息学软件对SVA衣壳蛋白的理化性质、热点残基、相互作用力等进行分析。将SVA病毒液在不同温度(4、25、37℃)及不同pH值(4.0、5.8、6.6、7.2)缓冲液中进行孵育,利用TCID50和实时荧光定量PCR(RT-qPCR)对SVA进行稳定性分析。结果表明:SVA衣壳蛋白在特定条件下具有不稳定性且衣壳蛋白耐热性不高,蛋白质间通过弱相互作用结合而形成病毒颗粒。通过SVA热稳定和酸稳定的试验验证随孵育温度升高或pH值下降,SVA病毒颗粒稳定性显著下降。综上,SVA具有热不稳定性和酸不稳定性,为新型高效疫苗与热稳定保护剂的研发提供一定参考。 Senecavirus A(SVA),as one of the pathogens of swine vesicular infectious diseases,is affecting the swine industry in China.To explore the effects of temperature and acidic conditions on the replication of SVA,CHN/SVA/HB/2018 SVA strain was examined in the study.Bioinformatics software was employed to analyze the physical and chemical properties hotspot residues,and interaction forces of capsid proteins of SVA.In addition,SVA was incubated at different temperatures(4,25,37℃)or with different pH(4.0,5.8,6.6,7.2)buffers.TCID50 and RT-qPCR methods were used for the stability analysis of SVA.The results showed that SVA capsid proteins were unstable under certain conditions with low heat resistance.The virus particles were formed by weak interaction between the proteins.The stability of SVA virus particles decreased significantly with the increase of incubation temperature or the decrease of pH value.It is suggested that SVA has thermal instability and acid instability,which provides some reference for the research and development of new high-efficiency vaccines and heat-stable protective agents.
