海洋真菌Fusarium sp.HL21活性次级代谢产物研究

摘要 目的研究海洋真菌Fusarium sp.HL21的次级代谢产物及其抗菌活性。方法采用硅胶柱层析、Sephadex LH-20凝胶柱层析及半制备高效液相色谱等方法,对该菌株发酵产物分离纯化;后通过核磁共振波谱(NMR)和质谱(MS)等方法并比对相关文献,鉴定化合物结构;采用96孔板微量肉汤稀释法和孢子萌发法评价其抗细菌和抗真菌活性。结果从该菌株中分离鉴定了8个化合物,分别为plancyin G(1)、吲哚-3-乙醛酸甲酯(2)、吲哚-3-乙酸(3)、丁二酸二甲酯(4)、阿魏酸(5)、gibepyrone F(6)、6-((2S,3S)-2,3-二羟基-2-丁醇基)-3-甲基-2-吡喃酮(7)和6-((2R,3S)-2,3-二羟基-2-丁醇基)-3-甲基-2-吡喃酮(8)。结论化合物5、7和8对青枯劳尔菌BXL018、甘蓝黑腐病菌BLY013和辣椒青枯病菌BLY014表现出一定的抑菌活性。 Objective To investigate the secondary metabolites from a marine-derived fungus Fusarium sp.HL21 and their antifungal and antibacterial activities.Methods Silica gel chromatography,Sephadex LH-20 gel column chromatography and Reversed-Phase High Performance Liquid Chromatography(RP HPLC)were used to isolate and purify the secondary metabolites from the fermented crude extract of the fungus Fusarium sp.HL21.The structures of the isolated compounds were elucidated by comparing the NMR and MS spectroscopic data with those of literatures.The antibacterial and antifungal activities were evaluated by the 96-well plate broth microdilution method and spore germination assay,respectively.Results Eight compounds were isolated and identified as plancyin G(1),(1H-indol-3-yl)oxoacetic acid methyl ester(2),3-indole acetic acid(3),diethyl succinate(4),ferulic acid(5),gibepyrone F(6),6-((2S,3S)-2,3-dihydroxybutan-2-yl)-3-methyl-2H-pyran-2-one(7)and 6-((2R,3S)-2,3-dihydroxybutan-2-yl)-3-methyl-2H-pyran-2-one(8).Conclusion Compounds 5,7 and 8 exhibited moderate antibacterial activities against Ralstonia solanacearum BXL018,Xanthomonas campestris pv.campestris BLY013 and Ralstonia solanacearum BLY014.
