黄连标准汤剂、配方颗粒与盐酸小檗碱对大肠埃希菌的抑菌作用机制研究 被引量:4

摘要 目的探究黄连标准汤剂、配方颗粒与盐酸小檗碱对大肠埃希菌的抑菌作用机制,为黄连配方颗粒抑菌机制的研究提供参考。方法利用二倍稀释法测定黄连标准汤剂、配方颗粒及盐酸小檗碱对大肠埃希菌(EC)的最小抑菌浓度(MIC)、最小杀菌浓度(MBC),并根据测定的结果进行EC抑菌曲线的绘制、胞外可溶性蛋白相对含量、电导率的研究。结果黄连标准汤剂、配方颗粒与盐酸小檗碱对EC的MIC分别为3.14、5.06和0.80 mg/mL;MBC的结果分别为3.14、10.13和3.19mg/mL;EC在黄连标准汤剂、配方颗粒与盐酸小檗碱质量浓度为1×MIC和1/2MIC的情况下生长受到明显抑制,空白组EC生长良好;在供试品质量浓度为1×MIC和2×MIC情况下,随着时间增加,EC胞外可溶性蛋白相对含量、电导率都有不同程度的上升。结论在黄连标准汤剂、配方颗粒与盐酸小檗碱的作用下,EC细胞膜通透性增加,使细胞内容物泄露,从而达到抑菌作用。 Objective To explore the antibacterial mechanism of Huanglian standard decoction,formula granules,and berberine hydrochloride on Escherichia coli,and to provide a reference for the study of antibacterial mechanism of Huanglian formula granules.Methods The minimum inhibitory concentration(MIC)and the minimum bactericidal concentration(MBC)of Coptis standard decoction,formula granules,and berberine hydrochloride on Escherichia coli(EC)were determined by the principle of the double dilution method.According to the results of the determination,the antibacterial curve of EC,the relative content of extracellular soluble proteins,and the conductivity were studied.Results The MICs of Huanglian standard decoction,formula granules,and berberine hydrochloride against E.coli were 3.14,5.06 and 0.80 mg/mL,respectively.MBC results were 3.14,10.13 and 3.19 mg/mL,respectively;the growth of EC was significantly inhibited when the mass concentrations of Huanglian standard decoction,formula granules,and berberine hydrochloride were 1×MIC and 1/2MIC,and the blank group grew well.Under the conditions of 1×MIC and 2×MIC,the relative content and conductivity of EC extracellular soluble proteins increased to varying degrees with the increase of time.Conclusion Under the action of Huanglian standard decoction,formula granules,and berberine hydrochloride,the permeability of EC cell membrane increases,and the cell content leaked,so as to achieve antibacterial effects.
