FoxO转录因子参与糖尿病心肌病脂代谢研究进展

摘要 糖尿病性心肌病(diabetic cardiomyopathy,DCM)是糖尿病心血管并发症发病和死亡的主要原因,脂代谢异常是导致DCM纤维化进而引起舒张功能障碍的重要原因。叉头框转录因子O(Forkhead transcription factor O,FoxO)在糖尿病早期心肌脂代谢紊乱中承担重要角色。FoxO通过转录代谢相关靶基因参与糖尿病早期糖异生、胰岛素抵抗、心肌脂代谢紊乱以及由代谢异常引发的氧化应激与炎症。本文重点阐述FoxO(主要是FoxO1)参与糖尿病心肌脂代谢过程,为预防治疗糖尿病心血管并发症提供新的角度。 Diabetic cardiomyopathy(DCM)is the primary cause of cardiovascular complications and death in individuals with diabetes mellitus.Abnormal lipid metabolism plays a significant role in the development of myocardial fibrosis and diastolic dysfunction in DCM.Recent studies have demonstrated the important role of Forkhead Transcription Factor O(FoxO)in early diabetic myocardial lipid metabolism disorders.FoxO is involved in transcriptional regulation of metabolism-related target genes,contributing to early diabetic gluconeogenesis,insulin resistance,myocardial lipid metabolism disorders,as well as oxidative stress and inflammation induced by metabolic abnormalities.This article focused on the involvement of FoxO(mainly FoxO1)in diabetic myocardial lipid metabolism,providing new insights for the prevention and treatment of diabetic cardiovascular complications.
